特异分子标记开发方法及其在杂交狼尾草真实性鉴定的应用

摘要 为了开发一种快速、准确鉴定杂交狼尾草真实性的鉴定方法,基于珍珠粟和象草基因组中的特异序列开发分子标记用于杂交狼尾草真实性鉴定,并通过对16份候选后代进行染色体计数和基因组原位杂交加以验证。结明表明:通过对珍珠粟A亚基因组、象草A'和B亚基因组进行比较序列分析,分别筛选出1146、2421、4067条特异序列,从这3个亚基因组中开发了21个分子标记,并最终筛选出3个高效的标记。PCR及琼脂糖凝胶电泳结果显示,16份候选后代中有13份为真实的异源三倍体杂交狼尾草,而闽牧6号、热研4号和桂牧1号被鉴定为四倍体象草亲本而非真实杂交狼尾草。染色体计数和GISH结果与分子标记完全一致,证实了该标记系统的可靠性。该方法成功建立了杂交狼尾草真实性鉴定技术,并为其他复杂多倍体植物杂交种的鉴定研究提供借鉴。 In order to develop a rapid and accurate identification method for the authenticity of hybrid Pennisetum,themolecular markers were developed based on the specific sequences derived from the genomes of pearl millet(Pennisetum glaucum)and elephant grass(Pennisetum purpureum)for the authenticity identification of hybridPennisetum,with validation conducted via chromosome counting and genomic in situ hybridization(GISH)on 16candidate progeny lines.The results showed that 1146,2421,and 4067 specific sequences were screened out by thecomparative sequence analysis of A subgenome of pearl millet,along with the A'and B subgenomes of elephant grass.A total of 21 molecular markers were developed from these three subgenomes,and three highly efficient markers wereultimately screened.The results of PCR amplification and agarose gel electrophoresis revealed that 13 out of the 16candidate lines were true allotriploid hybrid Pennisetum,while the lines Minmu No.6,Reyan No.4,and Guimu No.1were identified as tetraploid elephant grass rather than genuine hybrids.The results of chromosome counting andGISH were fully consistent with the molecular markers,which confirmed the reliability of the marker system.Thismethod successfully established the authenticity identification technology of hybrid Pennisetum,and provided areference for the identification of hybrids in other complex polyploid plant species.
