摘要
白豆杉(Pseudotaxus chienii)为中国特有的红豆杉科白豆杉属植物.为筛选可在白豆杉不同组织中稳定表达的内参基因,基于已有的白豆杉转录组数据,以白豆杉不同组织(根、茎、老叶、新叶)为材料,进行实时荧光定量PCR检测,并利用geNorm、NormFinder、BestKeeper、比较ΔCt法和RefFinder 5种方法,对6个候选内参基因B1、B2、B3、B4、B5和B6进行稳定性分析.结果表明,B3在白豆杉不同组织中表达最稳定,而B2表达最不稳定.因此,B3可作为白豆杉功能基因表达分析的最佳内参基因.
Pseudotaxus chienii,a monotypic genus within the Taxaceae family endemic to China,is a rare and protected species.To identify suitable reference genes for gene expression analysis in different tissues of P.chienii,this study selected six candidate reference genes(B1-B6)based on existing transcriptome data.Their expression stability was evaluated across various tissues(root,stem,old leaf,and new leaf)using real-time fluorescent quantitative PCR(RT-qPCR)and analyzed with five methods:geNorm,NormFinder,BestKeeper,comparativeΔCt method,and RefFinder.The results indicated that B3 was the most stably expressed gene across all tested tissues,whereas B2 was the least stable.Thus,B3 is recommended as the optimal reference gene for future functional gene expression studies in P.chienii.
