3个不同产地种质甘草中8种主要活性成分合成累积动态变化规律的研究

Study on the dynamic changes of synthesis and accumulation of 8 main active ingredients in glycyrrhiza germplasm from 3 different germplasm sources

摘要 [目的]以中国甘草道地产区杭锦旗、赤峰和会宁3个野生种质甘草引种于天津为研究内容,于1年生长期内每隔15 d采收1次样品,对所含8种主要功效成分含量进行测定,系统考察3个产地不同种质甘草不同采收期药材中活性成分合成累积规律及特点,筛选优良种源。[方法]半月采集1次样品,分别采用高效液相色谱(HPLC)法、NaNO2-Al(NO3)3-NaOH比色法、蒽酮-H2SO4法测定不同采收期样品中8种主要功效成分含量,考察功效成分合成累积动态变化趋势及规律。[结果]不同采收期3个种质甘草引种天津药材样品中所含甘草素、甘草次酸、异甘草素、总黄酮和总多糖合成累积量变化趋势基本一致,但会宁甘草中甘草苷、甘草酸和异甘草苷含量在不同采收时间内均高于杭锦旗和赤峰甘草。[结论]3个野生种质甘草引种天津后生长适应性良好,在1年生长周期内,不同采收期甘草主要活性功效物质合成功效成分动态合成量波动变化趋势差异并不显著,会宁种质甘草中功效成分合成累积量水平均高,具有较强的优良种质特性,因此,会宁甘草在天津盐碱土壤环境中规模化引种具有明显的潜力。 [Objective]The study focuses on the introduction of three wild varieties of Glycyrrhiza uralensis,Hangjin Banner,Chifeng,and Huining,from the licorice authentic production areas in China,into Tianjin.Samples were collected every 15 days during the one-year growth period to determine the content of eight main functional components.The synthesis and accumulation patterns and characteristics of active components in medicinal materials of different qualities and harvesting periods of licorice from the three production areas were systematically investigated,and excellent sources were screened.[Methods]Collect samples every half month,and use HPLC,NaNO2-Al(NO3)3-NaOH colorimetric method,and anthrone H2SO4 method to determine the content of 8 main active ingredients in samples at different harvesting periods.Investigate the dynamic changes and patterns of the synthesis and accumulation of active ingredients.[Results]The changes in the synthesis and accumulation of liquiritigenin,glycyrrhetinic acid,isoliquiritigenin,total flavonoids,and total polysaccharides in three germplasm licorice samples introduced to Tianjin at different harvesting periods were basically consistent.However,the content of liquiritin,glycyrrhetinic acid,and isoliquiritin Huining licorice was higher than that in Hangjinqi and Chifeng licorice at different harvesting times.[Conclusion]Three wild germplasm of Glycyrrhiza uralensis have good growth adaptability after being introduced to Tianjin.During the one-year growth cycle,there is no significant difference in the dynamic synthesis trend of the main active functional substances and functional components in licorice at different harvest periods.The Huining germplasm of licorice has a high level of cumulative synthesis of functional components and strong excellent germplasm characteristics.Therefore,the large-scale introduction of Huining licorice in Tianjin’s saline alkali soil environment has obvious potential.
