市场中驼乳粉的动物源性成分分析

摘要 目的分析市售驼乳粉的动物源性成分,评估其掺假风险。方法依据内蒙古地方标准[DB15/T 2026—2020《骆驼源性成分检测方法实时荧光PCR法》、DB15/T 2023—2020《牛和山羊源性成分同步检测方法实时荧光PCR法》],采用实时荧光聚合酶链式反应(polymerase chain reaction,PCR)技术检测20份驼乳粉样品中骆驼、牛及山羊源性成分,结合DNA浓度、纯度、循环(cycle threshold,Ct)值及扩增曲线综合分析检测结果。结果所有样品均检出骆驼源性成分(Ct值为16.25~24.14),15份样品检出牛源性成分(Ct值为15.85~24.51),未检出山羊源性成分;内源质控扩增稳定(Ct值为13.81~20.52),扩增曲线均呈典型S型,避免了假阴性结果,证实实验体系灵敏可靠。结论20份市售驼乳粉中均检出骆驼源性成分,部分样品同时检出牛源性成分,未检出山羊源性成分,实验体系灵敏可靠。实时荧光PCR技术可有效鉴别复杂基质中的动物源性成分,为驼乳粉质量监控与标识规范化提供技术支撑。 Objective To analyze the animal-derived materials of commercially available camel milk powder,and assess its risk of adulteration.Methods According to the local standards of Inner Mongolia(DB15/T 2026—2020 Authentification of camel derived materials-Real-time PCR method,DB15/T 2023—2020 Authentification of cattle and goat derived materials-Real-time PCR method),real-time fluorescent polymerase chain reaction(PCR)technology was used to detect the camel,cattle and goat derived materials in 20 camel milk powder samples.The results were comprehensively analyzed in combination with DNA concentration,purity,cycle threshold(Ct)value and amplification curve.Results Camel-derived materials were detected in all samples(Ct values were 16.25-24.14),cattle-derived materials were detected in 15 samples(Ct values were 15.85-24.51),and goat-derived materials were not detected.The endogenous control amplification was stable(Ct values were 13.81-20.52),and the amplification curves were all typical S-shaped,avoiding false negative results and confirming that the experimental system was sensitive and reliable.Conclusion Camel-derived materials have been detected in all 20 commercially available camel milk powder,some samples have also tested positive for cattle-derived materials,while no goat-derived materials have been detected.The experimental system has proven sensitive and reliable.Real-time PCR technology can effectively identify animal-derived materials in complex matrix,which provides technical support for quality monitoring and labeling standardization of camel milk powder.
