摘要
试验旨在分离海洋来源嗜盐芽孢杆菌B7的抗菌肽,并探究其理化性质。通过硫酸铵盐析、凝胶过滤层析纯化获得目标抗菌肽,分析其理化性质、抗菌谱及作用机制。结果显示,嗜盐芽孢杆菌B7抗菌肽具有广泛的抗菌活性,对多种动物病原菌具有显著的抑制作用。嗜盐芽孢杆菌B7抗菌肽在pH值4.5~10.5内表现出较强的抑菌活性,100℃加热处理20 min后仍保留25%的活性;对胰蛋白酶和胃蛋白酶具有较高的耐受性,但对蛋白酶K较为敏感;对恶臭假单胞菌和金黄色葡萄球菌的最小抑菌浓度分别为12.89和206.14 mg/L。透射电镜结果显示,嗜盐芽孢杆菌B7抗菌肽可破坏细菌的细胞壁和细胞膜的完整性,导致内容物泄漏。研究表明,海洋来源嗜盐芽孢杆菌B7抗菌肽的广谱性、pH值适应性、耐热性和蛋白酶耐受性符合饲料工业的需求。
The experiment aimed to isolate antimicrobial peptides from marine derived Bacillus halotolerans B7 and explore their physicochemical properties.The target antibacterial peptide was purified by ammonium sulfate salting out and gel filtration chromatography,and its physical and chemical properties,antibacterial spectrum and action mechanism were analyzed.The results showed that the Bacillus halotolerans B7 antimicrobial peptides exhibits broad-spectrum antimicrobial activity,showing significant inhibition against multiple animal pathogens.The Bacillus halotolerans B7 peptide maintained strong activity within a pH value range of 4.0 to 10.5 and retained 25%of its activity after heat treatment at 100℃for 20 minutes.It displayed high tolerance to trypsin and pepsin but was sensitive to proteinase K.The minimum inhibitory concentrations against Pseudomonas putida and Staphylococcus aureus were 12.89 and 206.14 mg/L,respectively.Transmission electron microscopy result showed that the antimicrobial peptides disrupted bacterial cell wall and membrane integrity,leading to cytoplasmic leakage.The study indicates that the broad-spectrum,pH adaptability,heat resistance and protease tolerance of the marine Bacillus halotolerans B7 antimicrobial peptide meet the requirements of the feed industry.
