基于UPLC-Q-TOF-MS/MS与定量指纹图谱的陈皮(樟头红)质量分析 被引量:1

Quality Analysis of Citri Reticulatae Pericarpium Based on UPLC-Q-TOF-MS/MS and Quantitative Fingerprinting

摘要 目的:基于超高效液相色谱-四极杆飞行时间质谱法(UPLC-Q-TOF-MS/MS)和定量指纹图谱技术,分析陈皮(樟头红Citrus reticulata‘Zhangshuensis’)的质量。方法:通过对18批陈皮(樟头红)药材进行检测,获得陈皮(樟头红)药材的对照指纹图谱,并采用UPLC-Q-TOF-MS/MS对标定的陈皮(樟头红)共有峰进行鉴定。采用相似度分析、聚类分析、正交偏最小二乘法-判别分析对陈皮(樟头红)进行质量分析与评价。结果:18批陈皮(樟头红)的相似度为0.988~0.999。在陈皮(樟头红)药材对照图谱中共标定24个共有峰,其中共鉴定和推测出21个成分,包括10个多甲氧基黄酮类化合物、4个黄酮氧苷类化合物、2个黄酮碳苷类化合物、3个苯丙素类化合物、2个柠檬苦素类化合物。结合化学模式识别技术,共筛选出陈皮(樟头红)与其他栽培变种陈皮的差异性质量标志物2个,分别为橘皮素、5-去甲基川陈皮素,实现了陈皮(樟头红)与其他变种陈皮的区分。结论:建立了陈皮(樟头红)的定量指纹图谱方法并对其共有峰进行鉴定,所建立的方法稳定可靠,可以实现陈皮(樟头红)与其他栽培变种陈皮的区分。研究结果可为陈皮(樟头红)的质量控制、质量评价及鉴别提供参考。 Objective:To investigate the quality of Citri Reticulatae Pericarpium produced from Citrus reticulata'Zhangshuensis'based on quantitative fingerprinting and ultra-performance liquid chromatography-quadrupole-time of flighttandem mass spectrometry(UPLC-Q-TOF-MS/MS).Methods:The fingerprints of 18 batches of Citri Reticulatae Pericarpium('Zhangshuensis')samples were obtained,and UPLC-Q-TOF-MS/MS was employed to identify the common peaks.The similarity analysis,cluster analysis,and orthogonal partial least squares-discriminant analysis were conducted to evaluate the quality of'Zhangshuensis'.Results:The fingerprint similarity of the 18 batches of samples ranged from 0.988 to 0.999.A total of 24 common peaks were identified in the reference fingerprint of'Zhangshuensis',among which 21 components were identified,including 10 polymethoxyflavonoids,4 flavone O-glycosides,2 flavone C-glycosides,3 phenylpropanoids,and 2 limonoids.On this basis,the chemical pattern recognition screened out two differential quality markers:hesperetin and 5-demethylnobiletin,which can distinguish'Zhangshuensis'from other varieties.Conclusion:This study establishes a quantitative fingerprinting method and identifies the common peaks of C.reticulata'Zhangshuensis'for the first time.This established method is stable and reliable and can distinguish'Zhangshuensis'from other cultivated varieties.The results can provide data reference and a scientific basis for the quality control,quality evaluation,and identification of'Zhangshuensis'.
