重楼皂苷Ⅰ调控PI3K/AKT信号通路抑制肝癌细胞增殖、侵袭及迁移的作用机制研究 被引量:8

摘要 目的 探讨重楼皂苷I(Polyphyllin I,PPI)对人肝癌细胞HCCLM3增殖、迁移、侵袭的影响及其机制研究。方法 CCK8方法检测PPI对HCCLM3细胞活力抑制作用;克隆形成实验观察PPI和Sorafenib(Sora)对HCCLM3细胞增殖能力影响;细胞划痕实验验证PPI和Sora对HCCLM3细胞迁移能力影响;Transwell实验观察PPI和Sora对HCCLM3细胞侵袭能力影响;Western Blot实验检测PPI和Sora对HCCLM3肝癌细胞MMP2、MMP9蛋白以PI3K/AKT信号通路标志蛋白影响。结果 CCK8实验结果显示,与对照组比较,不同浓度的PPI对HCCLM3细胞活力有明显的抑制作用,且呈时间和剂量依赖性(P<0.05);克隆结果表明,与空白对照组比较,PPI和Sora对HCCLM3细胞增殖能力有明显抑制作用,PPI联合Sora组抑制作用最明显(P<0.01);细胞划痕实验与Transwell结果显示,PPI和Sora对HCCLM3细胞迁移和侵袭能力也具有抑制作用,联合组给药组效果更好(P<0.01);Western Blot实验结果表明PPI和Sora对HCCLM3细胞转移标志蛋白有明显抑制作用,Sora联合PPI组的抑制效果更好(P<0.01)。结论 PPI可通过调控PI3K/Akt信号通路影响HCCLM3细胞的增殖、侵袭和迁移能力。 Objective To investigate the effect and mechanism of Polyphyllin I(PPI)on the proliferation,invasion and migration of hepatocellular carcinoma.Methods The CCK8 assay was used to detect the cytotoxicity of PPI on HCCLM3 cells.The clone formation assay was confirmed the effect of PPI or Sorafenib(Sora)on the proliferation ability in HCCLM3 cells.The scratch-wound healing assay and Transwell assay were used to determine the influence of PPI or Sora on the migration ability in HCCLM3 cells.Western Blot experiment was used to ensure the mechanism of PPI or Sora in liver cancer cells.Results The results of CCK8 showed that different concentrations of PPI had obvious inhibitory effects on HCCLM3 compared with the control group,and there was a time-and concentration-dependent manner(P<0.05).The cloning results showed that compared with the blank control group,PPI and Sora had a significant inhibitory effect on the proliferation ability of HCCLM3 cells,and the PPI combined with Sora group had the most significant inhibitory effect(P<0.01).The cell scratch experiment and Transwell results showed that PPI and Sora also had inhibitory effects on the migration and invasion ability of HCCLM3 cells,and the combined group had better effects(P<0.01).The Western Blot experiment results showed that PPI and Sora had a significant inhibitory effect on HCCLM3 cell metastasis marker proteins,and the Sora combined with PPI group had a better inhibitory effect(P<0.01).Conclusion PPI may affect the power of proliferation,migration and invasion in HCCLM3 cells by regulating PI3K/Akt signaling pathway.
