知母多糖的提取工艺优化及抗氧化活性研究 被引量:2

Optimizing Extraction Process of Anemarrhena Rhizome Polysaccharide and Its Antioxidation Activity

摘要 目的研究知母多糖的超声波辅助碱液最佳提取工艺及其抗氧化活性。方法以知母多糖得率为指标,通过单因素实验,以超声功率、提取时间、料液比和碱液浓度为考察因素,结合Box-Behnken实验,利用响应面法优化超声波辅助碱液提取工艺。通过对1,1-二苯基-2-三硝基苯肼(1,1-diphenyl-2-picrylhydrazyl,DPPH)的清除率来评价知母多糖的抗氧化能力。结果当超声功率为92 W,提取时间为20 min,料液比为1∶40,碱液浓度为0.05 mol/L时,知母多糖的提取率可达34.059%。抗氧化活性实验结果显示,纯化的知母多糖具有较强的DPPH自由基清除能力,在质量浓度为1.772 mg/mL时,其对DPPH自由基的清除率高达88.11%。结论通过响应面优化所得提取工艺具有提取率高,稳定的优点,可用于知母多糖的提取;知母多糖具有较强的体外抗氧化活性。 Objective To study the ultrasonic-assisted extraction technology of Anemarrhena Rhizome polysaccharide and their antioxidant activity.Methods The polysaccharide was extracted by ultrasonic extraction method.On the basis of single factor ex⁃periment,Box-Behnken central composite experiment was used to establish quadratic regression equation with ultrasonic power,extraction time,solid-liquid ratio and lye concentration as the influencing factors and the yield of Anemarrhena Rhizome polysac⁃charide as the response value.The single factor response surface experiment was used to optimize the extraction process.The an⁃tioxidant activity of Anemarrhena Rhizome polysaccharide on 1,1-diphenyl-2-Picrylhydrazyl(DPPH)was studied.Re⁃sults When the ultrasonic power was 92 W,the extraction time was 20 min,the ratio of solid to liquid was 1∶40 and the concen⁃tration of lye was 0.05 mol/L,the extraction rate of Anemarrhena Rhizome polysaccharide reached 34.059%.The results of an⁃tioxidant activity experiment showed that the purified Anemarrhena Rhizome polysaccharide had strong DPPH free radical scaven⁃ging ability,and the DPPH free radical scavenging rate was 88.11%when the mass concentration was 1.772 mg/mL.Conclusion The optimized extraction process has the advantages of high extraction rate and stability,and can be used for the extraction of Anemarrhena Rhizome polysaccharide.Anemarrhena Rhizome polysaccharide has a good antioxidant activity in vitro.
