摘要
本文旨在研究circ RNA-0028171对三氧化二砷(As_(2)O_(3))诱导的血管内皮细胞凋亡的调控作用。用0~15μmol/L As_(2)O_(3)作用人脐静脉血管内皮细胞(human umbilical vein endothelial cells,HUVECs)24 h,用MTT法检测HUVECs损伤情况,用实时定量PCR检测circ RNA-0028171及凋亡相关蛋白Bcl-2、Bax m RNA表达水平,用Western blot检测Bcl-2、Bax蛋白表达水平。通过转染circ RNA-0028171过表达质粒和si RNA观察circ RNA-0028171是否参与As_(2)O_(3)对HUVECs的调控。结果显示,与对照组相比,As_(2)O_(3)组细胞活性显著下降,Bcl-2/Bax m RNA比值和蛋白比值显著降低,circ RNA-0028171显著低表达。过表达circ RNA-0028171抑制As_(2)O_(3)诱导的HUVECs凋亡,而敲减circ RNA-0028171促进As_(2)O_(3)诱导的HUVECs凋亡。上述结果提示,circ RNA-0028171参与调控As_(2)O_(3)诱导的血管内皮细胞凋亡过程。
The present study was aimed to investigate the effects of circ RNA-0028171 on the apoptosis of vascular endothelial cells induced by arsenic trioxide(As_(2)O_(3)).Human umbilical vein endothelial cells(HUVECs)were treated with 0–15μmol/L As_(2)O_(3)for 24 h.Then,cellular viability was measured by MTT assay.The expression levels of circ RNA-0028171,Bcl-2 and Bax m RNA were detected by real-time quantitative PCR.Bcl-2/Bax protein ratio was detected by Western blot.Whether circ RNA-0028171 was involved in the regulation of HUVECs by As_(2)O_(3)was investigated by transfection with overexpression plasmid of circ RNA-0028171 and si RNA.The results showed that compared with the control group,As_(2)O_(3)group showed decreased cellular viability,reduced Bcl-2/Bax m RNA and protein ratios,and significantly lower expression of circ RNA-0028171.Overexpression of circ RNA-0028171 inhibited apoptosis of HUVECs induced by As_(2)O_(3).Knockdown of circ RNA-0028171 by si RNA promoted As_(2)O_(3)-induced apoptosis in HUVECs.These results suggest that circ RNA-0028171 is involved in the vascular endothelial cell apoptosis induced by As_(2)O_(3).
