ICAM-1、MMP-9及NF-κB在胆管癌中的表达及与外周血免疫抑制细胞的关系

Expressions of ICAM-1,MMP-9 and NF-κB in cholangiocarcinoma and their correlation with immunosuppressive cells in peripheral blood

摘要 目的分析细胞间黏附分子-1(ICAM-1)、基质金属蛋白酶-9(MMP-9)及核转录因子(NF-κB)在胆管癌中的表达及与外周血免疫抑制细胞的关系。方法选取2019年12月至2020年12月收治的60例胆管癌患者作为胆管癌组,同期选取60例体检的健康人作为健康组。采用酶联免疫分析法(ELISA)和免疫组化检测胆管癌组、癌旁组ICAM-1、MMP-9、NF-κB表达,流式细胞仪检测胆管癌组、健康组外周血免疫抑制细胞内MDSC表达情况,分析ICAM-1、MMP-9、NF-κB在胆管癌中的表达及与外周血免疫抑制细胞的相关性。结果与癌旁组织相比,癌组织中ICAM-1、MMP-9、NF-κB表达升高(P<0.05);ICAM-1、MMP-9、NF-κB表达水平胆管癌患者肿瘤直径无关,与TNM分期、分化程度、淋巴结转移、浸润深度表达相关,Ⅲ期、Ⅳ期、中低分化、有淋巴结转移、≥1/2肌层浸润胆管癌患者ICAM-1、MMP-9、NF-κB表达升高(P<0.05);胆管癌组外周血中MDSC比例高于健康组,差异有统计学意义(P<0.05);ICAM-1、MMP-9、NF-κB、外周血免疫抑制细胞之间相关性分析显示,ICAM-1、MDSC之间呈正相关(r=0.597,P=0.001),MMP-9、MDSC之间呈正相关(r=0.585,P=0.001),NF-κB、MDSC之间呈正相关(r=0.596,P=0.001)。结论在胆管癌中ICAM-1、MMP-9、NF-κB表达水平升高,外周血免疫抑制细胞比例升高,且ICAM-1、MMP-9、NF-κB指标与外周血免疫抑制细胞相关,临床上可根据其表达预测此病。 Objective To investigate the expressions of intercellular adhesion molecule-1(ICAM-1),matrix metalloproteinase-9(MMP-9)and nuclear transcription factor(NF-κB)in cholangiocarcinoma and their correlation with immunosuppressive cells in peripheral blood.Methods Sixty patients with cholangiocarcinoma who were treated in our hospital from December 2019 to December 2020 were enrolled as cholangiocarcinoma group,and 60 healthy subjects who underwent physical examination in our hospital were enrolled as control group.The Enzyme-linked immunoassay(ELISA)and immunohistochemistry were used to detect the expression levels of ICAM-1,MMP-9,NF-κB in the cholangiocarcinoma group and the paracarcinoma group,and the expression levels of internal MDSC expression in immunosuppressive cells were detected by flow cytometry.Moreover the correlation between the expressions of ICAM-1,MMP-9,NF-κB and peripheral blood immunosuppressive cells was analyzed.Results Compared with those in para-cancerous tissues,the expression levels of ICAM-1,MMP-9,and NF-κB in cancer tissues were significantly increased(P<0.05),which were not related to the tumor diameter of patients,however,which were related to TNM staging,degree of differentiation,lymph node metastasis,deep infiltration expression.The expression levels of ICAM-1,MMP-9,NF-κB in the patients at stageⅢ,Ⅳstage,with moderately poorly differentiation,lymph node metastasis,≥1/2 muscular infiltration were significantly increased(P<0.05),and the proportion of MDSC in cholangiocarcinoma group was significantly higher than that in control group(P<0.05).Moreover there was a positive correlation between ICAM-1 and MMP-9,between MMP-9 and MDSC,and between NF-κB and MDSC(P<0.01).Conclusion The expression levels of ICAM-1,MMP-9,and NF-κB in patients with cholangiocarcinoma are increase,and the proportion of peripheral blood immunosuppressive cells is increased,and the expressions of ICAM-1,MMP-9,NF-κB are correlated with peripheral blood immunosuppressive cells,and the disease can be predicted according to the expression levels.
