摘要
目的:对二妙散水提液的急性毒性试验研究及安全性评价。方法:预试验:分别灌胃给予昆明小鼠二妙散水提液,剂量为209.04 g·kg^(-1)、156.78 g·kg^(-1)、130.65 g·kg^(-1)、104.52 g·kg^(-1),计算半数致死量(LD_(50))。最大耐受量试验:灌胃给予小鼠水提液剂量为130.65 g·kg^(-1),观察给药后小鼠的状态和死亡情况。给药14 d后检测小鼠的血常规、血液生化指标、主要脏器系数、体质量以及主要脏器病理学观察。结果:未能测出二妙散水提液的半数致死量。最大耐受量试验结果显示,实验小鼠给药14 d后状态良好,未见中毒死亡情况。第7、14天体质量增加与对照组比较无显著差异(P>0.05);实验小鼠血常规、血生化指标及主要脏器系数与正常对照组相比均差异无统计学意义(P>0.05);病理形态学检查未发现二妙散水提液对小鼠主要脏器产生病理改变。结论:二妙散水提液的最大耐受量为130.65 g·kg^(-1),超过该剂量会产生毒性作用。
Objective:To evaluate the acute toxicity of Ermiao Powder water extract and its safety.Methods:In terms of the pilot test,the median lethal dose(LD_(50))was measured after the administration of Ermiao Powder water extract with the doses of 209.04 g·kg^(-1),156.78 g·kg^(-1),130.65 g·kg^(-1) and 104.52 g·kg^(-1) in mice.In terms of maximum tolerance test,the state and death of the mice were observed after the administration of Ermiao Powder water extract at the dose of 130.65 g·kg^(-1).Blood routine examination,blood biochemical indicators,main organ coefficients and body mass were detected,and main organ pathological changes were observed after 14 days of administration in mice.Results:The 50%lethal dose of Ermiao Powder water extract could not be measured.The experimental results of the maximum tolerance showed that the experimental mice were in good condition,and there was no poisoning death after 14 days of administration.There was no significant difference in the increase of body mass on the 7th and 14th days compared with that in the control group(P>0.05).There were no significant differences in blood routine examination,blood biochemical indexes and main organ indexes compared with those in the blank control group(P>0.05).Pathological examination did not show that Ermiao Powder water extract had pathological changes on the main organs of mice.Conclusion:The maximum tolerance dose of Ermiao Powder water extract is 130.65 g·kg^(-1),exceeding which will cause toxic effects.
