黄龙止咳颗粒对咳嗽变异性哮喘大鼠气道高反应、气道炎症及气道重塑的干预效应 被引量:23

Effects of Huanglong Zhike Granule on Airway Hyperresponsiveness,Airway Inflammation and Airway Remodeling in Cough Variant Asthma Rats

摘要 目的:观察黄龙止咳颗粒对咳嗽变异性哮喘(cough variant asthma,CVA)大鼠气道高反应、气道炎症及气道重塑的干预效应。方法:卵清蛋白诱导CVA模型,按随机数字表法分为空白组、模型组、中药组、西药组及联合组,药物干预14 d后观察大鼠气道高反应,支气管肺泡灌洗液(bronchoalveolar lavage fluid,BALF)炎症细胞计数,肺组织形态学改变,血清及BALF中白介素4(Interleukin-4,IL-4),γ干扰素(Interferon-γ,IFN-γ),转化生长因子β(transforming growth factor-β,TGF-β)含量。结果:(1)模型组气道阻力明显高于空白组(P<0.01),各治疗组气道阻力较模型组降低(P<0.05),联合组最显著(P<0.01);(2)模型组BALF中炎症细胞较空白组增加(P<0.01),各治疗组炎症细胞较模型组降低(P<0.05),联合组最显著(P<0.01);(3)模型组肺组织可见炎症细胞浸润,杯状细胞增生,上皮下胶原沉积,各治疗组上述改变较模型组减轻,联合组最显著。(4)与空白组相比,模型组血清及BALF中IL-4、TGF-β含量增加(p<0.01),IFN-γ含量降低(P<0.01);与模型组比较,各治疗组血清及BALF中IL-4、TGF-β含量降低(P<0.05),IFN-γ含量增加(P<0.05),联合组最显著(P<0.05)。结论:黄龙止咳颗粒可降低CVA大鼠气道高反应,减轻气道炎症,延缓气道重塑,其机制可能与调节细胞失衡、减少TGF-β含量相关。其对CVA的疗效与激素相当,可作为激素的替代用药,若与激素合用疗效更优。 Objective:To investigate the effect of Huanglong Zhike on airway hyperresponsiveness,airway inflammation and airway remodeling in cough variant asthma(CVA)rats.Methods:CVA models were induced by ovalbumin,and randomly divided into blank group,model group,traditional Chinese medicine group,western medicine group and combined group.Parameters including airway hyperresponsiveness,inflammation cells in bronchoalveolar lavage fluid(BALF)and lung histomorphology were measured after intervention for 14 days,plasma and BALF levels of interleukin-4(IL-4),interferon-γ(IFN-γ),transforming growth factor-β(TGF-β)were also tested.Results:(1)The airway resistance in model group was higher compared with blank group(P<0.01),which was significant reduced by different treatments(P<0.05).The combined group had the lowest airway resistance among all treatment groups(P<0.01);(2)The number of inflammation cells in model group were increased compared with blank group(P<0.01),that were decreased in all treatment groups compared with model group(P<0.05).The combined group had the least inflammatory cells among treatment groups(P<0.01);(3)Inflammatory cell infiltration,goblet cell hyperplasia and subepithelial collagen deposition were observed in lung tissue of model group.Those changes were suppressed in treatment groups;(4)The plasm and BALF levels of IL-4,TGF-βwere increased in the model group compared with the blank group(P<0.01).In contrast,levels of IFN-γwere decreased(P<0.05).Levels of IL-4 and TGF-βwere decreased in all treatment groups compared with model group(P<0.05),as well as IFN-γwere increased(P<0.05).Conclusions:Huanglong Zhike,Which would be can be used as a replacement drug for hormones,could suppress airway hyperresponsiveness,airway inflammation and airway remodeling of CVA,and the effects could enlarge if combined treated with glucocorticoid.The underline mechanism may be related to regulate the immune imbalance and reduction of TGF-β.
