摘要
本研究采用脂多糖(LPS)诱导细胞因子建立小鼠脑水肿模型,评估不同作用时间(2、4、12、24 h)甘露醇对小鼠脑水肿的治疗效果。观察实验前后脑水含量差异,测定细胞因子含量(IL-1β、AQP4、TNF-α),再研究脑组织光学内源信号变化规律,最后基于这些参数分析甘露醇对脂多糖致小鼠脑水肿治疗效果。LPS组脑含水量低于NS组(P<0.05),MT组脑水含量较LPS组均显著升高(P<0.05);LPS组三种因子含量较NS组明显增加(P<0.05),MT组较LPS组均明显减少(P<0.05);血氧参数HbO2、Hb与细胞因子AQP4均具有高度相关性(0.8<|r|<1)。研究表明,AQP4与HbO2、Hb分别呈高度负相关、高度正相关,且不同作用时间MT组对小鼠脑水肿治疗均有效果,24 h组最明显。
We used lipopolysaccharide(LPS)induced cytokines to establish mouse brain edema model and to evaluate the therapeutic effects of mannitol on brain edema in mice at different time points(2,4,12,24 h).The differences of brain water contents before and after the experiment were observed,the levels of cytokines(IL-1β,AQP4,TNF-α)were measured,the changes of optical endogenous signal in brain tissue were studied.Finally,based on these parameters,the therapeutic effect of mannitol on brain edema induced by lipopolysaccharide was analyzed.Brain water content in LPS group was lower than that in NS group(P<0.05),and brain water content in MT group was significantly higher than that in LPS group(P<0.05);the contents of three factors in LPS group were significantly higher than that in NS group(P<0.05),while that in MT group was significantly lower than that in LPS group(P<0.05);blood oxygen parameters HBO2,Hb and cytokine AQP4 were highly correlated(0.8<|r|<1).The results show that AQP4 is highly negatively correlated with HbO2 and positively correlated with Hb.The effect of MT group on brain edema is obvious in 24 h group.
