摘要
【目的】旨在评估饲料铜水平对黄鳝肝脏生化指标及肝脏、肠道组织结构的影响。【方法】以五水硫酸铜(CuSO4·5H2O)为铜源,按等对数间距梯度配置了5组(CL1、CL2、CL3、CL4、CL5)饲料,各组饲料Cu2+含量测定分别为14.21,23.95,37.01,135.63,499.63 mg/kg,饲喂均质量为(62.09±1.07)g的黄鳝60 d后取样,分析黄鳝肝脏生化指标、观察肝脏及肠道组织结构。【结果】①CL3、CL4及CL5组肝脏抗氧化能力显著高于CL1及CL2组,CL4及CL5组肝脏脂质过氧化程度显著高于CL1、CL2及CL3组;②CL1、CL2及CL3组黄鳝肝细胞完整,线粒体及内质网结构清晰、分布规律;CL4及CL5组肝细胞肿胀、空泡化,细胞核偏移于细胞一侧,内质网肿胀断裂、线粒体破碎凋亡;③相较前3组,CL4及CL5组肠道绒毛损伤更为严重,肠绒毛杯状细胞数量急剧增加。【结论】饲料中补充适量的铜能够提升黄鳝肝脏抗氧化能力,但过量添加会加重肝脏脂质过氧化程度。黄鳝可通过增加肠绒毛杯状细胞数量方式促进肠黏液分泌,从而增强对饲料铜的屏障能力,但这种屏障能力有限,高铜饲料最终导致黄鳝肝脏结构受损,肝细胞内的线粒体及内质网破碎凋亡,胞内大量脂滴沉积。
[Objective]The aim of this study was to evaluate the effects of dietary copper level on liver biochemical indexes and liver and intestinal histological structure of swamp eels.[Method]In the experiment,copper pentahydrate sulfate(CuSO4·5H2O)was used as the copper source,and five groups(CL1,CL2,CL3,CL4and CL5)feeds were configured by adopting the method of equal logarithmic spacing(LOG10)gradient.The Cu2+content of each group was 14.21,23.95,37.01,135.63,499.63 mg/kg,respectively.The swamp eels(62.09±1.07)g were sampled 60 days after feeding.The effects of dietary copper level on the liver biochemical indexes,and intestinal and liver histological structure of swamp eels were analyzed[.Result]The results showed that:(1)The antioxidant capacity of liver in CL3,CL4 and CL5 groups was significantly higher than that in CL1 and CL2 groups,and the degree of lipid peroxidation of liver in CL4 and CL5 groups was significantly higher than that in CL1,CL2 and CL3 groups.(2)In CL1,CL2 and CL3 groups,eel hepatocytes were intact,mitochondria and endoplasmic reticulum were clearly structured and distributed.In CL4 and CL5 groups,hepatocytes were swollen and vacuolated,the nucleus was shifted to one side of the cell,the endoplasmic reticulum was swollen and broken,and mitochondria were broken and apoptotic.(3)Compared to the first three groups,CL4 and CL5 groups had more serious intestinal villi damage,and the number of intestinal villi goblet cells increased sharply.[Conclusion]The results showed that appropriate copper supplementation in the feed could improve the antioxidant capacity of swamp eel liver,but excessive addition could aggravate the degree of lipid peroxidation in the liver.Swamp eel can promote intestinal mucous secretion by increasing the number of intestinal villus goblet cells,thus enhancing the barrier ability from dietary copper.However,this barrier ability is limited,and the high copper feed eventually leads to the damage of eel’s liver structure and the fragmentation of mitochondria and endoplasmic reticulum in hepatocytes,and the deposition of a large number of lipid droplets.
