摘要
本实验旨在探究ADIPOQ基因在贵州黑山羊不同组织中的表达谱和在不同月龄脂肪组织中的表达规律。采用qRT-PCR检测了贵州黑山羊8个组织样(心、肝、脾、肺、肾、胃、背最长肌和脂肪组织)及5个不同时期(1、3、6、9、12月龄)脂肪组织中ADIPOQ基因的表达量。结果表明:ADIPOQ基因在黑山羊8个组织中均有表达,其中在脂肪组织的表达量最高,肾脏中表达量最低;时序表达结果表明ADIPOQ基因总体表达趋势为先增加再减少,其中在6月龄表达量最高,在12月龄表达量最低。说明ADIPOQ基因对贵州黑山羊脂肪代谢具有一定的调控作用,或可作为研究黑山羊肉质品质的候选基因。
The purpose of this study was to study the expression profile of ADIPOQ gene in different tissues of guizhou black goat and the expression rule in adipose tissues of black goat at different age stages.In this experiment,qRT-PCR was used to detect the mRNA expression of ADIPOQ gene in 8 tissue samples of Guizhou black goat,and to detect the expression regularity of ADIPOQ gene in adipose tissue of black goat at 5 different time periods.The results showed that ADIPOQ gene was expressed in all tissues tested by black goat,and the expression level in adipose tissue was the highest,and the expression level in kidney was the lowest.Sequential expression results in adipose tissue showed that the overall expression trend of ADIPOQ gene increased first and then decreased,with the highest expression at the age of 6 months and the lowest expression at the age of 12 months.This indicates that ADIPOQ gene has a certain regulatory effect on the metabolism of adipose tissue in Guizhou black goat,or can be used as a candidate gene for studying the meat quality of black goat.
