摘要
目的研究LncRNA NEAT1在胃癌中的临床意义。方法采用荧光定量PCR法检测胃癌组织及癌旁正常组织标本中LncRNA NEAT1表达水平,分析NEAT1的表达水平与胃癌患者临床病理学参数及预后的相关性。结果NEAT1在胃癌组织中的表达高于癌旁正常组织(P<0.01)。NEAT1的表达与胃癌患者脉管侵犯、远处转移、以及是否复发显著相关(P<0.05),而与患者年龄、性别、分化程度、神经侵犯、分期、淋巴结转移无显著相关性。单因素与多因素COX回归分析显示,NEAT1表达水平为患者无病生存期及总体生存期相关,且可以作为胃癌患者预后的独立危险因素;生存分析结果显示,与NEAT1低表达组相比,NEAT1高表达组胃癌患者预后较差。结论NEAT1可能在胃癌中发挥着促癌作用,可作为临床诊断及评估患者预后的潜在指标。
Objective To study the clinical significance of LncRNA NEAT1 in gastric cancer.Methods The expression level of LncRNA NEAT1 in gastric cancer tissues and adjacent normal tissues was detected by fluorescence quantitative PCR,and the correlation between the expression level of NEAT1 and the clinicopathological parameters and prognosis of gastric cancer patients was further analyzed.Results The expression of NEAT1 in gastric cancer tissues was higher than that in normal tissues(P<0.01).The expression of NEAT1 was significantly correlated with vascular invasion,distant metastasis,and recurrence in patients with gastric cancer(P<0.05),but not with age,gender,degree of differentiation,nerve invasion,staging,and lymph node metastasis.Univariate and multivariate COX regression analysis showed that the expression level of NEAT1 was related to disease-free survival and overall survival of patients,and could be used as an independent risk factor for the prognosis of gastric cancer patients.Survival analysis showed that gastric cancer patients in the NEAT1 group with high expression had a poor prognosis.Conclusion NEAT1 may play a role in promoting cancer in gastric cancer,and can be used as a potential indicator for clinical diagnosis and prognosis evaluation.
