霉菌毒素吸附剂对玉米赤霉烯酮脱毒效果的评价 被引量:11

摘要 本试验旨在评价几种霉菌毒素吸附剂对玉米赤霉烯酮(ZEA)的体外和体内脱毒效果。分别采用pH 3.0和6.5的缓冲盐溶液为反应介质,分析比较市场上常见的12种霉菌毒素吸附剂对ZEA的体外吸附能力,依据分析结果选取体外吸附效果最好的3种吸附剂A3、A4和A6进行动物试验评价。选取健康三元杂交(杜×长×大)青年母猪40头,按日龄[(75±2)日龄]和平均体重[(29.19±0.76)kg]相近原则分为5组,每组8个重复,每个重复1头。对照组饲喂不含ZEA的基础饲粮,ZEA组饲喂添加1 mg/kg ZEA的基础饲粮,试验E1、E2、E3组饲喂在ZEA组饲粮基础上分别添加质量分数为0.2%的吸附剂A3、A4和A6的饲粮。预试期7 d,正试期21 d。通过比较不同组间猪只的生长性能、阴户宽度、器官指数、血清生化指标和血浆中ZEA含量及其代谢产物的残留水平评价吸附剂的体内脱毒效果。结果表明:1)12种霉菌毒素吸附剂在不同pH反应体系中对ZEA的体外吸附效果不同,其中吸附剂A3、A4和A6对ZEA的体外吸附效果最佳,在pH 3.0和6.5的反应介质中吸附率均达到90%以上。2)与对照组相比,饲粮中添加1 mg/kg的ZEA对青年母猪的平均日增重、平均日采食量、料重比,肝脏指数、脾脏指数、肾脏指数,血清中总蛋白、白蛋白、免疫球蛋白G含量及碱性磷酸酶(ALP)、谷氨酸脱氢酶(GLDH)、谷草转氨酶(AST)、谷氨酰转肽酶(GGT)和肌酸激酶(CK)活性均没有显著影响(P>0.05),但能显著增加阴户宽度、生殖器官指数、血浆中ZEA含量及其代谢物α-玉米赤霉烯醇(α ZOL)和β-玉米赤霉烯醇(β ZOL)含量(P<0.05),并显著降低血清中雌二醇含量(P<0.05)。3)与ZEA组相比,饲粮中添加0.2%的吸附剂A4和A6能使生殖器官指数恢复至对照组水平。3种吸附剂对血清中雌二醇的含量均有一定恢复效果。吸附剂A4能显著改善阴户红肿并降低血浆中ZEA及其代谢物的含量(P<0.05)。由此可见,市场上不同霉菌毒素吸附剂对ZEA的体外吸附效果差异较大,体外吸附效果并不能完全代表体内脱毒效果,霉菌毒素吸附剂的脱毒效果评价需要体内与体外试验相结合进行。 This study was conducted to assess the in vitro and in vivo detoxification effects of different myco toxin absorbents on zearalenone(ZEA).In vitro binding efficacy of ZEA by twelve different mycotoxin ab sorbents in buffer solutions with pH 3.0 and 6.5 was analyzed and compared.The absorbents A3,A4 and A6 with the highest in vitro binding efficacy were selected for in vivo evaluation.Forty healthy prepubertal gilts(Duroc×Landrace×Large white)were allocated into 5 groups according to age[(75±2)days of age]and av erage body weight[(29.19±0.76)kg](8 replicates per group and 1 pig per replicate).Prepubertal gilts in control group and ZEA group were fed a basal diet supplemented with 0 and 1 mg/kg ZEA,respectively.The experimental groups(E1,E2 and E3)were fed the ZEA group diet supplemented with 0.2%(w/w)different absorbents(A3,A4 and A6).The adaptation period lasted for 7 days and the trial period lasted for 21 days.The in vivo detoxification effect of adsorbents was evaluated by comparing the growth performance,vulva width,organ indexes,serum biochemical indexes and the contents of ZEA and its metabolites in plasma among different groups.The results showed as follows:1)in vitro adsorption test revealed that 12 kinds of adsorbents had different adsorption capacity in buffer solutions with different pH,of which A3,A4 and A6 had the high est binding efficacy on ZEA,and the adsorption rates reached more than 90%in both media with pH 3.0 and 6.5.2)Compared with control group,average daily feed intake,average daily gain,feed to gain ratio,liver index,spleen index,kidney index,the contents of total protein,albumin,immunoglobulin G and the activities of alkaline phosphatase,glutamate dehydrogenase,glutamic oxalacetic transaminase,glutamyltranspeptidase and creatine jubase in serum of prepubertal gilts were not significantly affected in ZEA group with the addition of 1 mg/kg ZEA(P>0.05).However,vulva width,reproductive organ index and the contents of ZEA,α zearalanel(ZOL)andβ ZOL in plasma were significantly increased(P<0.05),while the estradiol content in serum was significantly decreased(P<0.05).3)The addition of 0.2%absorbent A4 or A6 in ZEA diet could recover the reproductive organ index to the level of control group.All the three adsorbents recovered the estra diol content in serum to some degree.Adsorbent A4 could significantly decreased vulva width,the contents of ZEA and its metabolites in plasma(P<0.05).It is concluded that different mycotoxin absorbents has different adsorption capacity on ZEA in vitro,and the in vitro adsorption capacity cannot completely represent the in vi vo detoxification effect.The evaluation of the detoxification effect of mycotoxin adsorbents requires the combi nation of in vivo and in vitro trials.[Chinese Journal of Animal Nutrition,2020,32(5):2116 2125]
