摘要
目的研究前列腺癌组织中跨膜丝氨酸蛋白酶2-E26转录因子(TMPRSS2-ETS)融合基因及ETS相关基因(ERG)、ETS变异体1(ETV1)、同源丢失性磷酸酶张力蛋白基因(PTEN)、雄激素受体(AR)蛋白表达情况。方法收集大同市第三人民医院2016年12月至2017年10月前列腺癌患者石蜡标本50例,以同期前列腺增生患者标本30例作为对照。采用荧光原位杂交检测TMPRSS2-ERG、TMPRSS2-ETV1以及TMPRSS2-ETV4基因融合状况;免疫组织化学法检测ERG、ETV1、PTEN、AR蛋白的表达。结果前列腺癌组织中TMPRSS2-ETS融合基因阳性率为70%(35/50),前列腺增生组织中未检测到TMPRSS2-ETS融合基因。TMPRSS2-ETS融合基因的表达在不同年龄、血清前列腺特异性抗原(PSA)值及是否远处转移前列腺癌患者组间的差异均无统计学意义(均P>0.05),而Gleason评分>7分的前列腺癌患者TMPRSS2-ETS融合基因的表达高于≤7分的患者,差异有统计学意义(P<0.05)。ERG及ETV1在前列腺癌组织中的表达高于前列腺增生组织,差异均有统计学意义(均P<0.05);PTEN在前列腺癌组织中的表达低于前列腺增生组织,差异有统计学意义(P<0.05);AR在前列腺癌组织及前列腺增生组织中的表达差异无统计学意义(P>0.05)。TMPRSS2-ETS融合基因与ERG在前列腺癌中的表达呈正相关(r=0.302,P<0.05);TMPRSS2-ETS融合基因与ETV1、PTEN、AR在前列腺癌中的表达均无相关性(均P>0.05)。结论TMPRSS2-ETS融合基因和ERG、ETV1蛋白在前列腺癌中表达上调,可能参与了肿瘤的发生、发展,检测TMPRSS2-ETS融合基因和ERG蛋白有助于前列腺癌的诊断。
Objective To study the expressions of transmembrane protease serine 2-E26 transformation specific (TMPRSS2-ETS) fusion gene and ETS related gene (ERG), ETS variant 1 (ETV1), phosphatase and tensin homolog deleted on chromosome 10 (PTEN), androgen receptor (AR) in prostate cancer. Methods A total of 50 cases of prostate cancer paraffin specimens and 30 cases of benign prostatic hyperplasia as the controls in the same period from the Third People's Hospital of Datong between December 2016 and October 2017 were collected. TMPRSS2-ERG, TMPRSS2-ETV1 and TMPRSS2-ETV4 gene fusion status were detected by using fluorescence in situ hybridization. Immunohistochemistry was used to detect the expressions of ERG, ETV1, PTEN and AR protein. Results In prostate cancer tissues, TMPRSS2-ETS fusion gene positive rate was 70%(35/50), and TMPRSS2-ETS fusion gene was not detected in benign prostatic hyperplasia. The expression of TMPRSS2-ETS fusion gene in prostate cancer patients with different age, serum prostate specific antigen level and whether distant metastasis had no statistically significant differences (all P > 0.05). The expression of TMPRSS2-ETS fusion gene in Gleason score > 7 patients was higher than that in Gleason score≤7 patients (P < 0.05). The expressions of ERG and ETV1 in prostate cancer tissues were higher than those in benign prostatic hyperplasia, and the differences were statistically significant (both P < 0.05). The expression of PTEN in prostate cancer tissues was lower than that in benign prostatic hyperplasia (P < 0.05). There was no significant difference in the expression of AR between prostate cancer and benign prostatic hyperplasia tissues (P > 0.05). The expressions of TMPRSS2-ETS fusion gene and ERG in prostate cancer were positively correlated (r=0.302, P < 0.05), and there was no correlation between the expression of TMPRSS2-ETS fusion gene and the expressions of ETV1, PTEN and AR (all P > 0.05). Conclusions The expressions of TMPRSS2-ETS fusion gene and ERG, ETV1 protein are upregulated in prostate cancer, which may involve in the occurrence and development of prostate cancer. The detection of TMPRSS2-ETS fusion genes and ERG protein can provide a reference for the diagnosis of prostate cancer.
