摘要
为了证实大肠杆菌活菌数与其吸光度之间的关系,本次试验分别接种10 uL、20 uL、30 uL、40 uL、50 uL至8 mL培养液中培养8 h,分别使用无菌营养肉汤与生理盐水调零测定其OD_(600)值,倍比稀释8次,使用平板菌落计数法测定菌液中的活菌数量。结果表明,大肠杆菌菌液OD_(600)值与其活菌数成线性关系,使用生理盐水调零时回归方程为:y=475.98x-132.93(R^2=0.9973);使用无菌营养肉汤调零时回归方程为:y=475.98x-149.59 (R^2=0.9973)。大肠杆菌在培养至第8 h时,菌液中活菌数与其OD_(600)值呈良好线性关系,分光光度法可以用于大肠杆菌活菌的计数。
The purpose of this paper was to verify the mathematical relationship between the number of live E.coli and its absorbance.In this study,E.coli was respectively inocuLated into 10 uL,20 uL,30 uL,40 uL and 50 uL nutrient solution and cuLtivated for 8 hours.After that,using sterile nutritious broth normal saline as zero adjustment,the OD600 value of them was measured.Then the bacteria solutions were respectively diluted 8 times and the number of living bacteria couLd be determined by the plate colony counting method.The resuLt showed that the OD600 value of the bacteria solution had a favorable linear relationship with the number of living bacteria.The regression equation was y=475.98x-132.93(R^2=0.9973)when using normal saline to zero it,while it was y=475.98x-149.59(R^2=0.9973)when using sterile nutrient broth to zero it.When it was cuLtivated for 8h,it was concluded that the number of living bacteria had a good linear relations with the value of OD600,which referred UV-spectrophotometry can be applied to the counting of Escherichia coli.
