摘要
本文旨在探讨芒果苷对高糖诱导系膜细胞增殖的抑制作用,并从自噬角度阐述可能的机制。采用CCK8法检测芒果苷对高糖诱导系膜细胞增殖的抑制作用,Western blot检测芒果苷给药24 h后LC3-II蛋白的表达,采用GFP-LC3绿色荧光视踪检测芒果苷对系膜细胞自噬体数量的影响。结果表明:与高糖组相比,芒果苷5×10^(-5)、1×10^(-5)、5×10^(-6)mol/L能明显改善系膜细胞增殖(P<0.01)。与高糖组相比,芒果苷给药24 h后5×10^(-5)、1×10^(-5)mol/L组LC3-II蛋白的表达升高(P<0.05)。芒果苷给药组(1×10^(-5)、2×10^(-5)、4×10^(-5)、8×10^(-5)mol/L)1 h、3 h GFP-LC3绿色点状荧光明显增多。提示芒果苷具有抑制高糖诱导下系膜细胞的增殖作用,其机制可能是通过提高系膜细胞自噬水平,从而维持细胞内环境的稳定。
To study the anti-proliferative effect of mangiferin on mesangial cell with high glucose condition, and its role on mesangial cell autophagy. CCK8 method was used to detect the anti-proliferative effect of mangiferin on mesangial cell with high glucose condition. After treatment with 24 h, the expression levels of autophagy marker LC3-Ⅱ protein in these five groups were detected by western blot. The GFP-LC3 green fluorescent trace method was used to detect autophagy of mesangial cells. The results showed that compared with high glueose group, Mangiferin treatment group ( 5 × 105 , 1 × 10.5 ,5 × 10.6 mol/L) can significantly inhibit mesangial cell proliferation ( P 〈 0. 01 ). Compared with high glucose group, LC3-Ⅱ protein expression level of mangiferin treatment group (5 × 10-5 , 1 × 10.5 tool/L) was higher (P 〈0. 05 ). Mangiferin treatment group ( 1 × 10.5 , 2× 10.5 ,4 × 10.5 , 8 ×10.2 mol/L) , the green fluorescent dot significantly increased. Mangiferin can inhibit the proliferation of mesangial cells with high glucose condition, its mechanism may be through improving the autophagy level of mesangial cell, maintaining the stability of environment in mesangial cell.
