摘要
目的 观察五灵胶囊对小鼠免疫功能的影响 .方法 小鼠免疫器官质量法观察五灵胶囊对免疫器官质量的影响 ;溶血素测定法观察体液免疫功能 ;E花结与脾抗体生成细胞试验观察细胞免疫功能 ;以 LPS+BCG诱导小鼠免疫性肝损伤观察五灵胶囊对一氧化氮 (NO)、白细胞介素 2 (IL- 2 )、白细胞介素 6(IL - 6)的影响 ;戊巴比妥钠诱导小鼠 P450 观察五灵胶囊对小鼠 P450 含量和睡眠时间的影响 .结果 五灵胶囊能使免疫小鼠 E花结数、脾脏指数、胸腺指数和 QHS增加 ,促进体内 Ig G与 Ig M释放 ,血清 HCIg G与 HCIgm水平上升 .对免疫性肝损伤小鼠微粒体内 NO五灵胶囊大剂量有促进作用 ,小剂量有抑制作用 ,对 IL- 2与 IL- 6影响不显著 .戊巴比妥钠能诱导小鼠的 P450 含量增加 ,五灵胶囊并不影响 P450 含量 ,但却抑制 P450 对戊巴比妥钠的代谢 ,使睡眠时间延长 .此外 ,五灵胶囊还能降低戊巴比妥钠诱导升高的谷胱苷肽 S转移酶(GSH- ST) .结论 五灵胶囊是一种免疫增强剂 ,对体液免疫与细胞免疫均有促进作用 ,不诱导 P450 产生 ,但能抑制 P450 对戊巴比妥钠的代谢 ,对 LPS诱导的细胞因子 ,五灵胶囊作用不显著 ,对微粒体内 NO呈双向作用 .
AIM To investigate the effect of Wuling capsule on immune function of mice. METHODS The influence of Wuling capsule on the weighty immune organ was observed by the method of immune organ weighty, the function of hu moral immune by detection of hemolysin, and the impact of cell mediate immune by both test rosette formation and quantitative haemolysis of SRBC (QHS). After immunological liver injury in mice induced by micro lipopoly saccharide (LPS) and bacilli calmette Guein (BCG), the content of IL 2 and IL 6, and the content of nitric oxide (NO) of hepatic microsomes, were measured. The content P 450 of hepatic microsomes and sleep time of the mice were detected by injection of sodium pentobarbital. RESULTS Wuling capsule was an immunity excitation reagent, which could increase the index organ of spleen and thyme, the rosette formation, and elevated the level Hc IgG and Hc IgM . The big dosage of Wuling capsule promoted and small dosage inhibited the action on NO of hepatic microsomes, and it did not exert much effect on IL 2 and IL 6 of mice immune liver injuries induced by LPS, wuling capsule didn't add to the content of P450 for hepatic microsome but prolonged sleep time in pentobarbital mice, and it could reduce the glutasthine s transferase( GSH ST)in mice induced by pentobarbital as well.CONCLUSION Wuling capsules are an immunity enhancer, which promote cellular and humoral immunity without, increasing P450 of hepatic microsomes. They have no effects on liver cytokine induced by LPS, and have double action on NO of hepatic microsomes.
