角蛋白金属结合体的制备及清除超氧阴离子自由基性能

摘要 本文在制备可溶性羽毛角蛋白溶液的基础上,制备了羽毛角蛋白(FK)结合ZnⅡ、Cu^(Ⅱ)、Mn^(Ⅱ)和Ni^(Ⅱ)的角蛋白金属结合体(FKM,M=Zn,Cu,Mn,Ni)。采用IR、UV-Vis、TGA、CD和SEM等对其结构进行了表征和分析。用核黄素/蛋氨酸-NBT还原法考察了角蛋白金属结合体FKM清除超氧阴离子自由基(O_(2)·^(-))的性能,探讨了其清除机理。结果表明:含不同金属离子的结合体均具有清除O_(2)·^(-)的能力。其中,Cu结合体的抗氧化活性最强。 The water-soluble biopolymer,feather keratin-metal conjugates(FKM),were prepared by the soluble feather keratin(FK)conjugating with metal ion(Zn^(Ⅱ),Cu^(Ⅱ),Mn^(Ⅱ),Ni^(Ⅱ)).The structures of FKM were characterized by IR,UV-Vis,TGA,CD and SEM.The superoxide anion radical(O_(2)·^(-))scavenging activity of FKM was investigated by NBT photochemical reduction and the effect mechanism of scavenging O_(2)·^(-)was also explored.The results show that the conjugates with different metal ions had antioxidant activities.After combining with the biopolymer FK,the Cu conjugate(FKCu)shows the most excellent scavenging ability of superoxide anion radical(O_(2)·^(-)).
