摘要
目的:探讨小檗碱(Ber)对慢性脑缺血(CCI)大鼠是否具有保护作用。方法:将30只Sprague-Dawley大鼠随机分为假手术组(10只)、模型组(10只)和Ber治疗组(10只)。采用双侧颈总动脉永久性阻断法建立大鼠CCI模型,各组均于术后2 h开始,经腹腔注射Ber(30mg/kg/d)或等容积生理盐水,每日1次,连续给药60 d。用Morris水迷宫法测试大鼠学习记忆功能,HE染色法观察皮层下脑白质病理改变。结果:与假手术组比较,模型组和治疗组逃避潜伏期(EL)均明显延长(均P<0.01),穿越平台次数均明显减少(均P<0.01)。与模型组比较,Ber治疗组EL明显缩短(P<0.01),穿越平台次数明显增多(P<0.05)。病理检查结果显示,假手术组大鼠皮层下脑白质结构正常。模型组大鼠皮层下脑白质严重疏松,神经纤维排列紊乱,大量空泡样改变,大量固缩神经细胞。治疗组大鼠皮层下脑白质轻度疏松,神经纤维排列不整齐,少量空泡样改变,散在固缩神经细胞。结论:Ber能够减轻CCI大鼠脑白质损伤和认知功能障碍。
Objective To explore whether berberine(Ber)has protective effect against chronic cerebral ischemia(CCI)in rats.Methods 30 Sprague-Dawley rats were randomly divided into three groups including sham-operated group,model control group,and Ber-treated group.CCI model rats were induced by permanent occlusion of bilateral common carotids(2-VO).Rats were treated by intraperitoneal injection with Ber(30mg/kg/d)or the same volume of normal saline 2h after operation and with the same dosage once daily for 60 days.The cognitive function was assessed with Morris water maze,and the pathological changes in subcortical white matter were examined by HE staining.Results Comparing with sham-operated group,the escape latency(EL)significantly extended(all P〈0.01)and the times of passing through the platform remarkably decreased(all P〈0.01)in model group and Ber-treated group.Comparing with model group,the EL significantly shortened(P〈0.01)and the times of passing through the platform remarkably increased(P〈0.05)in Ber-treated group.Pathological examination showed normal white matter structure appearing in sham-operated group;serious leukoaraiosis,disorderly arrangement of nerve fibers,numerous cytoplasmic vacuoles,and abundant pyknosis neuron in subcortical white matter appearing in model group;mild leukoaraiosis,irregularly arrangement of nerve fibers,a small amount of cytoplasmic vacuoles,and scattered pyknosis neuron in subcortical white matter appearing in Ber-treated group.Conclusion Ber can attenuate cerebral white matter injury and cognitive impairment caused by CCI in rats.
