参麦汤对糖耐量低减大鼠脂联素、瘦素及脂联素受体R2mRNA表达的影响 被引量:4

Effect of Shenmai Decoction on Adiponectim,Leptin and Expression of Adiponectin Receptor R2mRNA of Impaired Glucose Tolerance Rat

摘要 目的:观察参麦汤对糖耐量低减(Impaired Glucoseolerance,IGT)大鼠脂联素(Adiponectim)、瘦素(leptin)及脂联素受体R2mRNA表达的影响。方法:高脂高糖饲料喂养结合腹腔注射小剂量链脲佐菌素溶液的方法复制糖耐量低减大鼠模型;用参麦汤治疗;实验结束后颈动脉取血,采用酶免法测血清Adiponectim及leptin,应用RT-PCR方法测定大鼠肝组织脂联素受体R2mRNA的表达。结果:和正常组比较,模型组Adiponectim浓度降低,有显著性差异(P<0.05),leptin的浓度升高,有极显著性差异(P<0.001);与模型组比较,各干预组均可提高Adiponectim浓度、降低leptin水平,其中以参麦汤加运动组为著(P<0.01),参麦汤组(P<0.05)、运动组(P<0.05)亦有明显差异;与正常组相比,模型组大鼠脂联素受体R2mRNA表达明显降低(P<0.001),各治疗组与模型组相比,参麦汤加运动组、参麦汤组大鼠脂联素受体R2mRNA表达上升显著(P<0.001),运动组(P<0.05)次之。结论:在IGT阶段已经出现leptin、Adiponectim分泌异常,参麦汤能显著地降低leptin水平,提高Adiponectim水平,促进脂代谢,上调糖耐量异常大鼠肝脂联素受体R2mRNA表达。推测参麦汤能改善机体能量的调节和维持代谢稳态。 Objective:To study the effect of Shenmai Decoction on Adiponectim,Leptin and expression of adiponectin receptor R2 mRNA of impaired glucose tolerance(IGT) rats.Methods:High fat and sugar feed combined with intraperitoneal injection of low dose of streptozotocin method were used for replicating impaired glucose tolerance rats model.After treatment with Shenmai Decoction,the levels of Adiponectim and Leptin were detected with enzyme-linked immune method.The liver expression of adiponectin receptor R2 mRNA was detected with RT-PCR.Results:Compared with the normal group,model group's adiponectim levels reduced and there were significant differences(P〈0.05) and leptin concentration increased and there was a very significant difference(P〈0.001).Compared with model group,the intervention group can improve the concentration of Adiponectim,lower levels of leptin and Shenmai Decoction and exercise group was the most obvious(P 〈0.01),and Shenmai Decoction group(P〈 0.05) and exercise group(P 〈 0.05) also had obvious difference.Compared with normal group,model group rats' R2 mRNA adiponectin receptors expression reduced significantly(P〈 0.001).The treatment group compared with model group,Shenmai Decoction and exercise group and Shenmai Decoction group's R2 mRNA adiponectin receptor expression in rats increased significantly(P 〈 0.001) and that of the exercise group was also significant(P〈 0.05).Conclusion:In the stage of IGT,there appeared abnormal secretion of Leptin and Adiponectim.Shenmai Decoction can significantly reduce Leptin levels,improve the level of Adiponectim,promote fat metabolism,increase impaired glucose tolerance in rats' liver R2 mRNA adiponectin receptor gene expression.Speculation and Shenmai Decoction can improve the body energy metabolism of adjusting and maintaining the steady state.
