摘要
于 1 997年 4月和 2 0 0 0年 5月对山东沿海海藻进行较系统的采集 ,对其中 39种海藻的 96个样品采用高通量方法进行选择性细胞毒筛选。结果表明 ,小粘膜藻 (Leathesiadif formes)、多管藻 (Polysiphoniaurcedata)、萱藻 (Scytosiphonlomentarius)、海萝 (Gloiopeliisfur cata)、叉开网翼藻 (Dictyopterisdivaricata)、点叶菜 (Punctarialatifolia)等对KB细胞或HT -2 9细胞具有选择性抑制活性。乙醇、氯仿的提取效率较高 ,多管藻的乙醇、氯仿提取物 ,萱藻的乙醇提取物和叉开网翼藻的正己烷提取物都有强的选择性细胞毒活性。
In order to study the antitumor activity of extracts from Shandong marine algae, 96 crude extracts of 39 species belonging to the classes Rhodophyceae, Chlorophyceae and Phaeophyceae were assayed for anti-KB and HT-29 activity. The algae were collected from the coastal areas of Qingdao and Weihai, Shandong Province in April 1997 and May 2000. Some kinds of seaweed showed potential antineoplastic activities, such as Leathesia difformes, Polysiphonia urcedata, Scytosiphon lomentarius, Gloiopeliis furcata, Dictyopteri divaricata, Punctaria latifolia, Symphyocladia latiuscula, Rhodomela confervoides, and Gracilaria vervucosa.;The extracts of Leathesia difformes have most potent cytotoxicity to KB cells (LD 50 =12.65μg/ml) and low activities to normal cells (LD 50 >50μg/ml). The cytotoxic activities were found prominent in the methanolic extracts of Polysiphonic urcedata (LD 50 =26.01μg/ml against Human Tumor-29). The other algae, Symphyocladia latiuscula, Rhodomela confervoides and Punctaria latifolia, had also shown marked inhibition of growth of KB cells, and the extracts of the Symphyocladia latiuscula, Rhodomela confervoides had shown selective activities against HT-29 cells.;Different extracting solvents of ten species of seaweed were each examined for their extracting efficiency. It turned out that ethanol was the best one, then chloroform, both of them were better than methanol. The extracts of ethanol and chloroform of Polysiphonia urcedata, the ethanolic extracts of Scytosiphon lomentarius and the hexane extracts of Dictyopteri divaricata have strong selective cytotoxic activities; all of the LD 50 against KB are less than 4.40μg/ml. The ethanolic extracts of Scytosiphon lomentarius have mighty activities against HT-29 cells (LD 50 =1.49μg/ml).;As all the algae grew broadly on the Shandong coast and their extracts inhibited KB or HT-29 tumor cells with a low side effect, they are promising sources for antitumor drugs.
