摘要
目的 :研究驻极体对 3T3细胞内游离 Ca2 +浓度变化的影响 ,探讨驻极体诱导 3T3细胞凋亡与游离 Ca2 +浓度变化之间的关系。方法 :选用 - 30 0 V和 - 5 0 0 V驻极体作用 3T3细胞 48h,即时依次加入 Ca Cl2 、A2 31 87、EDTA ,通过流式细胞术检测细胞内 Ca2 + 浓度的变化和 3T3细胞的凋亡量。结果 :(1)负极性驻极体作用 3T3细胞后 ,3T3细胞出现 10 %的凋亡量 ;(2 )驻极体作用 3T3细胞以后 ,细胞质游离 Ca2 +浓度升高 ,Ca2 +浓度的升高不是由于细胞内钙池的释放 ,而是由于细胞外 Ca2 +内流所引起。 结论 :驻极体诱导 3T3细胞凋亡的机制可能是细胞外 Ca2 + 内流引起的。
Objective: To study the changes of intracellular free Ca 2+ levels in 3T3 cells induced by electrets and discuss the relationship between electrets induced apoptosis of 3T3 cells and the changes of free Ca 2+ levels. Methods: After 48 h action of -300 V and -500 V electrets in 3T3 cells, CaCl 2, A 23187 , EDTA were added in proper order to 3T3 cells. The levels of intracellular Ca 2+ and the cell number of apoptosis were detected with flow cytometry. Results:(1)After the action of electrets, 10% of 3T3 cells appeared apoptosis. (2) After the action of electrets in 3T3 cells, the free Ca 2+ levels in cellular plasm were increased. The increasement of Ca 2+ levels was caused by the Ca 2+ inflow from outside cell, rather than the release of calcium pool inside cell. Conclusion:The electret can induce Ca 2+ outside 3T3 cell to flow into cells, thereby induce cell apoptosis.
