耳针改善血管性痴呆大鼠记忆与nNOS表达的关系 被引量:27

摘要 目的 探讨耳针对血管性痴呆大鼠 (VD)学习记忆的改善及与 n NOS蛋白表达的关系。 方法 采用 4-血管阻断的方法 ,建立大鼠 VD模型 ,然后进行耳穴肾、脑针刺治疗 ,用免疫组化法检测海马n NOS蛋白的变化 ;并用 Nissl染色方法 ,结合图象分析统计海马神经元丢失比率 ;同时采用 Y型迷宫 ,进行行为学检测 ,定量测定其学习记忆成绩。 结果 耳针治疗后 VD大鼠海马 CA1 区 n NOS蛋白表达减少 ,海马 CA1 区神经元丢失比率较 VD模型组减少 ,与学习记忆成绩呈负相关。 结论 耳针改善了VD大鼠学习记忆 ,这可能是针刺抑制脑缺血后 n NOS的过量增加 ,达到对 VD大鼠海马神经元的保护作用。 ObjectiveTo probe into the improvement of learning and memory of vascular dementia rats by ear acupuncture and its relati onship with expression of nNOS. MethodsVascular dementia rat model was established by blockin g 4 vessel, then the rats were treated by acupuncture at ear points kidney and Nao, the immunohistochemistry method was used to detect the nNOS protein, and th e lost rate of hippocampus neurons was calculated by Nissl staining method combi ned with image analysis. In the meantime, Y type maze was also used to measure the behavior of the rats.ResultsAfter acupuncture, the expression of nNOS in CA 1 area o f rat's hippocampus was reduced and the lost rate of neuron in CA 1 area of rat's h ippocampus is less than that in VD model group, which is negatively corrective w ith learning and memory.ConclusionsEar acupuncture could improve the learning and memory o f Vdrat and its mechanism may be that acupuncture could inhibit the over increa sed nNOS after cerebral ischemia to protect the hippocampus neuron of VD rat.
