摘要
以甘蓝型油菜 Brassica napus双低品种湘油 13号为材料 ,对其下胚轴再生频率影响因素诸如种子萌发培养基、外植体、基本培养基、激素、愈伤组织龄期和 Ag NO3浓度等进行了研究 .结果表明 ,在种子萌发培养基中加入适量激素 ,种苗质量好 ,其下胚轴愈伤组织发生早 ,形成快 ,诱导频率高 .选取 4d苗龄的种苗下胚轴靠近根基部或近子叶端 0 .3~ 0 .5 cm长为外植体 ,能使愈伤组织发生早 ,形成快 ,诱导频率也高 .以 B5 + 2 ,4- D0 .3m g/ L+ 6 - BA3mg/ L 为诱导培养基 ,MS+ 6 - BA3m g/ L+ GA30 .5 mg/ L 为分化培养基 ,最终诱导频率高 .在分化培养基中再加入适量 Ag NO3,芽苗分化频率成倍提高 ,以加入 2 0 μm ol/ L Ag NO3分化频率最高 .选取 16 d的愈伤组织 。
Factors affecting regeneration frequency of hypocotls of rapeseed Brassica napus . Xiangyou 13 such as germinating medium,explant,basic medium,hormone,age of callu,concentration of AgNO 3 and so on were studied. The results read as follows: the seedlings which germinated in the medium with proper hormone were good. The callus derived from their hypocotls initiated earlier,developed faster with a higher induced frequency. 0.3~0.5 cm,4 day hypocotls close to root or to cotyleden were better explants. The final induced frequency reached its highest point when using B 5 with 0.3 mg/L 2,4 D and 3 mg/L 6 BA as induced medium and MS with 3 mg/L 6 BA and 0.5 mg/L GA 3 as differentiated medium. When proper AgNO 3 was added into the differentiated medium,the differentiated frequency of callus would rise by times. Here 20 μmol/L AgNO 3 had the highest differented frequency. The differentiated effect of 16 day callus was good.
