摘要
根据鸡新城疫病毒 (NDV )、鸡传染性支气管炎病毒 (IBV)和鸡传染性喉气管炎病毒 (ILTV)的基因文库 ,设计了3对分别与NDV、IBV和ILTV某段基因序列互补的引物。用这 3对引物对同一样品中的NDV、IBV、ILTV核酸模板进行三重PCR扩增 ,结果均同时得到了 3条与设计相符的 310bp(NDV)、172 0bp(IBV)和 6 47bp(ILTV)三重PCR扩增带 ,而对其他 6种禽病病原的PCR扩增结果均为阴性 ;敏感性测定结果表明 ,该三重PCR技术能检出 10 pg的IBV、1pg的NDVRNA模板和 10 pg的ILTVDNA模板。
A triplex polymerase chain reaction was optimized to simultaneously detect three pathogens of NDV、IBV and ILTV in this article. three sets of specific primers were designed according to the sequences of NDV、IBV and ILTV at the GeneBank. It was shown the all samples which contained NDV、IBV cDNA and ILTV DNA,could be amplified by the triplex PCR using these three sets primers, yielding three specific bands of NDV 310 bp、IBV 1720 bp and ILTV 647 bp. But not specific band amplified from other six avian pathogenic virus and bacteria.As little as 10 pg of IBV、1 pg of NDV RNA and 10 pg of ILTV DNA were detected using gel electrophoresis in this triplex PCR.
