不同生长时期人参叶中几种水解酶的活力变化研究

摘要 目的:研究人参在生长季的不同时期叶中几种水解酶的活性变化。方法:以5年生人参叶为供试材料,采用中性磷酸缓冲溶液提取粗酶液。应用紫外分光光度法分别测定淀粉酶(AMY)、酯酶(EST)、酸性磷酸酯酶(ACP)、碱性磷酸酯酶(ALP)和植酸酶(PHY)的活力。结果:人参叶中AMY和EST活力在展叶期和结果期出现活力峰值,以展叶期活力最高;ACP、ALP和PHY活力以先上升后下降的趋势呈3个周期规律性变化,活力峰值分别出现在展叶期、结果期、果后参根生长期,其中ACP和ALP活力以展叶期活力最高,PHY以结果期活力最高。结论:在不同生长发育时期人参叶中的5种水解酶活力变化表现出阶段的特异性。 Objective: Researched at changes of hydrolase activity of ginseng leaves in the different growth stages of growing season. Methods: A five-year old ginseng leaves as material. Neutral phosphate buffer is selected to extract crude extract. UV was applied to determine the activities of amylase (AMY), esterase (EST), acid phosphatase (ACP), alkaline phosphatase (ALP), and phytase (PHY). Results: Activity peaks of AMY and EST appeared in the spreading stage and fruit growth stage, activity is highest in the spreading stage. Activity of ACP, ALP and PHY show regular changes of 3 cycles with first increased and then decreased. Activity peaks appeared in the spreading stage, fruit growth stage and the root growing after fruit stage. And activity of ACP andALP is highest in the spreading stage, activity of PHY is highest in the fruit growth stage. Conclusion: Activities of 5 kinds of hydrolase of ginseng leaves showed stage-specific changes in developmental growth stages.
