摘要
目的探讨Th17细胞在哮喘发病机制中的作用。方法以10例哮喘患者和10例健康志愿者为研究对象,分别留取外周血标本。利用RT-PCR技术检测外周血中淋巴细胞所表达的维甲酸相关较孤儿受体γt(RORγt)mRNA的相对量,流式细胞仪检测外周血中Th17细胞的比例,ELISA法检测血浆中白细胞介素(IL)-17的水平。结果 RORγt mRNA在哮喘组的表达水平(0.46±0.07)高于对照组(0.15±0.02),差异有统计学意义(P<0.01)。Th17细胞在哮喘组的比例(28.53±7.20)%高于对照组(14.72±2.33)%,差异有统计学意义(P<0.01)。IL-17在哮喘组的水平(102.31±11.45)pg/mL明显高于对照组(59.68±8.85)pg/mL。结论 Th17细胞与哮喘具有相关性,可能通过分泌以IL-17为主的细胞因子来上调炎症反应,加重哮喘病情。
Objective To explore the role of Th17 cells in the pathogenesis of asthma. Methods Peripheral blood was obtained from 10 patients with asthma(asthma group) and 10 healthy volunteers(control group). ROR~'t mRNA expression of tymphocytes was measured by RT-PCR, percentage of Thl7 cell was detected by FCM, IL-17 in plasma was examined by ELISA. Results ROR'/t mRNA level in asthma group(0.46±0.07) was significantly higher than that in control group(0.15±0.02) ,proportion of Th17 cell in asthma group(28.53± 7.20)M was significantly higher than that in control group( 14.72 ±2.33)%, P〈0.01. Com- pared with IL-17 level in control group(59.68±8.85)pg/mL, there was significant increase in asthma group(102.31 ± 11.45 )pg/ mL(P〈0.01). Conclusion Thl7 cell is associated with asthma,and probably aggravating asthma condition through increasing in- flammation secreting IL-17.
