摘要
酵母单杂交体系是借鉴酵母双杂交体系的基本原理 ,通过观察酵母细胞内报告基因的表达状况 ,研究DNA-蛋白质之间的相互作用。通过筛选 DNA文库可直接得到与靶序列相互作用的蛋白质基因序列 ,而无需分离、纯化蛋白 ,从而克服了体内研究的局限性。目前酵母单杂交体系已被广泛应用于寻找与目的 DNA片段相互作用的蛋白质分子 ,该体系是一种用于研究 DNA-蛋白质之间的相互作用的有效方法。本文就酵母单杂交体系的原理、具体实现方法、在研究 DNA结合蛋白质中的应用及其优缺点等方面作一简要介绍。
Yeast one hybrid system based on the principle of yeast two hybrid system, is applied for isolating novel genes encoding proteins that bind to a target, is acting regulatory element. The system may also be applied to map the DNA binding domain of previously known, or newly identified DNA binding proteins. Yeast one hybrid system offers maximal sensitivity because detection of the DNA protein interaction occurs while proteins are in their native configurations. In addition, the gene encoding the DNA binding proteins of interest is immediately available after a library screening. In this mini review, we introduce the principle, the experimental procedures, the applications, and the merits of yeast one hybrid system.
