摘要
经七步反应合成了Oxy-Cope重排反应的过渡态类似物-2-对羧丙氧基苯基环已醇(3),并以此为半抗原联接载体蛋白BSA制成抗原后免疫动物,从动物血清中分离纯化得到多克隆抗体。在200mmol/L的MES(4-吗啉乙磺酸)和200mmol/L的NaCl缓冲液中(pH=6.0),底物和抗体的摩尔比为100:1及37℃条件下,该抗体能催化3-对甲氧苯基-4-羟基-1,5-已二烯(1)重排生成6-对甲氧苯基-5-烯-已醛(2),v_(max)=15μmol/L·min,K_M=1025μmol/L,K_(cat.)=10min^(-1)。
A polyclonal antibody was generated against an antigen, a conjugate of carrier protein(BSA) and 2 -[ (p - (3 - carboxypropyloxy) - phenyl] - cyclohexanol - 1 (3) (a transition state analogue of Oxy - Cope rearrangement) , synthesized from cyclohexene in 7 steps. The antibody was found to catalyze the rearrangement of (35, 4R) -3 - p - methoxyphenyl - 4 - hydroxy -1,5- hexadiene (1) to trans - 6 - p - methoxyphenyl - 5 - hexe nal -1(2) with vmax = 15 μmol/L·min, KM = 1025 μmol/L and kcat.=10 min-1 in the presence of 0.2 mol/L MES [2- (N -Morpholino)ethanesulfonic acid] and 0.2 mol/L NaCl(pH = 6.0 buffer) at 37 ℃ with the mole ratio of (35, 4R) -3 - p - methoxyphenyl - 4 - hydroxy - 1,5 - hexadiene (1) to polyclonal antibody being 100:1.
