大鼠脑组织中丙戊酸的HPLC分析

摘要 用反相液相色谱 -柱前衍生法分析大鼠脑组织中的丙戊酸。采用脑组织碱化后水提 ,酸化后正己烷萃取的样品净化步骤 ,提高了分析灵敏度并减少了色谱干扰 ,其回收率平均为 80 5 %。脑组织中丙戊酸的线性范围为 0 1~ 2 0 μg/ 2 0 0mg ,最小检测量为 0 0 5 μg/ 2 0 0mg。最大RSD <5 74%。 A method has been developed for quantitating the antiepileptic drug valproic acid in rat brain by HPLC after percolumn derivatization.Studied extraction and chromatography needed to resolve analytes from a variety of endogenous constituents and to increase detection sensitivity.The separation was achieved by applying a reversed-phase C 18 column and methanol-water as mobile phase.The UV detector was set at 265 nm which allowed a linear range of determination 0 1~2 0 μg/200 mg of brain tissue and a sensitivity limit of detection 0 05 μg/200 mg.
