摘要
通过对不同分离株弓形虫抗原的分析 ,探讨弓形虫虫株间的差异性。取弓形虫 CAg阳性血液标本进行小鼠接种 ,对分离株进行生物学特性鉴定 ;采用流式细胞仪 ( FCM)对分离株弓形虫速殖子进行群体 DNA含量分析。结果在 5份 CAg阳性血液样本中分离到 4株弓形虫 ,两者符合率达 80 % ;经生物学鉴定结果显示 ,4个分离株的形态大小、毒力及抗原性均与 RH株弓形虫相似。上述 5株弓形虫速殖子 DNA含量分析均呈明显规律性 ,即均有 2个分布峰 ,但各虫株间的 2个分布峰中所占速殖子数存在显著性差异 ( P<0 .0 1)。表明检测弓形虫 CAg可作为弓形虫早期、急性期感染的确诊指标 ;采用流式细胞术进行 DNA群体分析 。
To observe the difference among Toxoplasma strains, we analyzed antigens of various strains of Toxoplasma. CAg positive blood was inoculated to mice, and then were biological feaures of isolatedstrains determinated, DNA contents of isolatedstrain populations were analyzed with Flow Cytometry(FCM). As a result, 4 strains of Toxoplasma were isolated from 5 CAg positive blood. The coincidence rate was 80%; The biological determination showed that 4 isolatedstrains were similar to RH in morphology, toxicity and antigen feature. Analysis of DNA content showed that the tachyzoites of above 5 strains of Toxoplasma all had two peaks, but significant difference in the number of tachyzoites of two peaks among strains was found. It is concluded that the detection of Toxoplasma CAg could make a definite diagnosis at the early and acute stage of Toxoplasma infection, analysis of DNA contents could reflect the difference of Toxoplasma strains.
