Progressive Atrophy of the Thalamus Following Middle Cerebral Artery Occlusion in Rats

摘要 目的:了解长期大脑中动脉闭塞(MCAO)后丘脑体积的改变。方法:35只大鼠分7组,除正常组和伪手术组外.其余各组用Tamura法闭塞大脑中动脉,用大鼠脑单克隆抗体微管蛋白-2(MAP-2)对各组大脑切片进行免疫组化染色.计算机测量各组大鼠脑切片的丘脑面积。结果:正常组0.99±0.01,伪手术对照2周组1.02±0.02,伪手术对照1个月组0.99±0.01,MCAO2周组0.87±0.03,1个月组0.78±0.03,MCAO3个月组0.71±0.04,6个月组0.61±0.05,伪手术组和MCAO组之间在丘脑萎缩方而有显著差异(P<0.001)。结论:提示逆行性退行性变在丘脑萎缩方而起重要作用。 Aim: To investigate the change of thalamus after long-term permanent middle cerebral artery occlusion (MCAO). Methods: 35 rats were divided into 7 groups in average, the middle cerebral artery was occluded with Tamura method except normal and sham groups. All rat slices were stained by the rat brain monoclonal antibody MAP-2 and the areas of the right and left thalamus were measured by computer digitizer. Results: The ration of the ipsilateral and contralateral tha- lamus was 0.99±0. 01 at normal group, 1.0±0.01 at sham group, 0.87±0. 03 at MCAO 2-week group, 0.78±0.03 at MCAO 1-month group, 0.71±0.04 at 3-month group, 0.61±0. 05 at 6-month group. There was a significant difference be- tween the sham group and MCAO groups (P<0.001). Conclusion: It is indicated that retrograde degeneration is thought to play an important role in thalamus atrophy.
