摘要
研究了采用高效液相色谱 紫外检测法对盐酸赖氨酸注射液中赖氨酸含量的测定 .作者在AminoQuant方法的基础上 ,改为仅采用邻苯二醛 (OPA)衍生化 ,省略了原方法衍生化试剂中的不稳定成分 ,而采用优化的反应条件提高紫外测定的灵敏度 ;又对这个组成较简单的样品改用恒定组成的流动相淋洗 ,手动方法测定避免了对该试样不必要的梯度淋洗 ,较原AminoQuant氨基酸分析法简便、快速 ,结果峰面积与浓度相关系数r =0 9998,回收率为 99 7% (RSD为 1 0 %~ 1 5 % ) .在采用紫外检测法时 ,最小检出量为 5 3× 10 -8(S/N =3)
Determination of lysine content in a lysine hydrochloride injection by the HPLC UV method was studied. The authors proposed to derivatize lysine with o phthalaldehyde(OPA)only, leaving out the unstable component(2 mercaptoethanol or 3 mercaptopropionic acid)in derivatizing reagent used in AminoQuant method for amino acid measurement. By optimizing reaction conditions, the sensitivity of lysine measurement can be increased significantly. Considering the simple composition of the sample, the authors proposed to analyze the derivatized sample with isocratic elution, simpler, manually operated instruments could thus be used for the measurement, thereby avoiding using gradient elution while unnecessary. The modified method was simpler and more rapid than the AminoQuant technique, whereas exact and precise results were obtained(linear correlation coefficient between peak area and concentration was 0 999 8, recovery 99 7%, within day and between day RSD was 1 0%~1 5%).If the UV detection was used, the sensitivity of the proposed method was found to be comparable with that by the AminoQuant method.
