摘要
目的 建立一种特异性强、灵敏度高的检测MN抗原的新方法。方法 应用抗M、抗N及抗血型糖蛋白A单克隆抗体 ,建立双位点一步ELISA法。结果 检测M、N抗原的最适pH值分别为 6 0及 7 5。适宜的反应温度为 2 0 30℃。利用表面活性剂l o n octyl β D glucopyranoside(OBG)浸泡血痕 ,可明显提高ELISA法检测陈旧血痕中N抗原的灵敏度 ,检测微量液体血和新鲜血痕的灵敏度分别为 0 .0 3μg血液和 0 .0 1~0 .0 5 μg干血。 结论 该法特异性强、灵敏度高 。
Objective To establish a new, specific and sensitive method for detecting MN antigen.Methods A one step sandwich ELISA method was established using anti M, anti N and anti glycoprotein A McAbs.Results The optimum pH value for detecting M and N antigens were 6.0 and 7.5 respectively, and the optimum temperatures was 20~30℃ . After blood stain was soaked with l o n ocytl β D glucopyranoside(OBG), a kind of surface active agent, the sensitivity of the one step sandwich ELISA method for detecting the N antigen in old blood stain was significantly improved. The sensitivities of the method for detecting microblood and fresh blood stain were 0.03 and 0.01 0.05μg of dry blood respectively.Conclusion The method shows strong specificity and high sensitivity. The study provides a new method for MN typing in forensic serology.
