摘要
以牛初乳为原料,采用离心方法进行脱脂,采用100 kD的平板膜超滤器进行超滤,探讨了牛初乳的纯化工艺,采用SDS-PAGE凝胶电泳检测超滤前后牛初乳蛋白的脱除情况。结果表明:牛初乳最佳脱脂工艺为:离心转速为3000 r/min,离心时间为4 0min,离心温度为25℃;最佳超滤纯化工艺为:超滤压力为0.25 MPa,超滤温度为40℃。经过超滤处理后杂蛋白明显减少,IgG含量明显增加。
Bovine colostrums were used as material. Fat in bovine colostnmas was removed by centrifugate. Ultrafilter with Plate membrane (100 kD, molecular, cut-off) was used to filter bovine colostrums. Purification technology of bovine colostrums was investigated. Removing of proteins before and after ultrafiltration in bovine colostrums was detected by SDS-PAGE gel electrophoresis. The results showed that the optimum technology of removing fat was as follows: centrifugate speed was 3 000 r/min, centrifugate time was 40 min, centrifugate temperature was 25℃. Purification technology of bovine colostrums by ultrafiltration was as follows: Ultrafiltration pressure was 0.25 MPa. Ultrafiltration temperature was 40 ℃. After ultrafiltration, Heteropolymeric proteins decreased obviously. Content of IgG increased obviously.
