香菇多糖、地黄煎影响小鼠免疫及内分泌功能的比较研究 被引量:15

Research on Comparison of Lentinan and Dihuang Decoction on Functions of Immune and Endocrine System of Mice

摘要 目的 :探讨香菇多糖与地黄煎对机体免疫及内分泌网络调节作用。方法 :采用KM雌性小白鼠随机分为4组:正常对照组、免疫低下模型组、免疫低下+香菇多糖(10 mg.kg-1)和免疫低下+地黄煎(10.4 g.kg-1)组。除正常对照组外,其余各组采用腹腔注射环磷酰胺造成小鼠免疫低下模型,各组分别按设计剂量连续给药1周。运用HE染色、酶联免疫吸附法(ELISA)等一系列方法观察小鼠卵巢和胸腺的形态变化,检测血清中IL-1α和雌二醇(E2)含量。结果 (:1)HE染色观察模型组胸腺狭窄细长,髓质明显萎缩,皮质较厚,生发中心减少,用药后均有不同程度的改善,其中以香菇多糖更为明显。模型组小鼠卵泡形状不规则,原始卵泡明显减少,闭锁卵泡增多,用药后可见多个闭锁卵泡和生长卵泡,其中地黄煎组可见多个原始卵泡;(2)ELISA检测血清中IL-1α含量模型组明显低于正常组,香菇多糖和地黄煎能不同程度提高IL-1α含量(免疫低下+香菇多糖组:175.81±46.00,免疫低下+地黄煎组:172.93±40.91),但明显低于正常组。E2含量模型组相比正常组并无明显降低,香菇多糖组与模型组比较无明显降低,而地黄煎组明显低于模型组。结论 :香菇多糖和地黄煎可不同程度提高小鼠免疫功能及调节内分泌的功能,各有偏颇。 Abstract: Ohjective: To investigate the adjusting effects of lentinan and Dihuang Decoction on the immune and endocrine functions. Methods: KM female mice were randomly divided into 4 groups: normal control group, immunocmnpromised model group, immunocompromised leutinan group and immunocompromised Dihuang Decoction group. Except normal control group, mice in other groups were all injected cyclophosphamidum ( CYC )into their abdomen to establish immunosuppression models. Drugs of respectively-designed doses were given intragastrieally tor one continuous week in each group. HE staining and enzyme linked immunosorbent assay ( ELISA ) were used to observe structural changes of thymuses and ovaries in mice and determine the contents of serum IL-1 ct and estrogen ( E2 ). Resuhs : In model groups, HE staining presented long, thin and small thymuses, conspicuously shrinking marrow, thicker cortex and noticeably decreased germinal centers. These symptoms were all improved after medication especially in lentinan group. Meanwhile, in model group, irregular shapes of fullicles, signally reducedprimordial follicles and increased atretic follicles were presenled on the owury lmthological secitons. Aftermedication, more atretic and growing follicles were observed. Furthernlore, in inmuanocompromised Dibtlang Deecoction group, more primordial follicles could be observed. In model group, level of serum inlterlukill 1 - α( 11,-1 α )was markedly decreased. Dihuang Decoction and lent inan both could increase IL-1α level toval.'ying degrees, but obviously lower than that in normal group. E2 level ill model group had no conspicuousdecrease. In eonlparison to model groups, E2 level in the immuno^ompromised Dihuang Decoction group significantly decreased, bill that of immunocompromised lentinan group didn't. Ctmt'hlsiot7 : Lentinan and Dbtaal^g Decoction can enhance lhe immune and endocrine tunc|ions of inuntmosuppressed mic.e at differentlevels with their own fimctional characteristics.
