输血相关急性肺损伤大鼠模型建立及肺组织病理研究 被引量:22

Pulmonary pathological changes in a rat model of transfusion-related acute lung injury

摘要 目的建立输注人类血浆大鼠输血相关急性肺损伤(TRALI)模型并分析其肺组织病理特点。方法将分离存储21 d后人AB型全血中的血浆,经静脉输注给经脂多糖预处理后的雄性Sprague Dawley大鼠,建立TRALI模型;分析大鼠肺组织病理及湿干比变化。结果输注了从存储后人全血中分离血浆的大鼠成功建立起TRALI模型,大鼠肺组织出现肺泡间隔增厚、肺泡内纤维蛋白浸润、肺泡内出血、支气管壁增厚等病理变化,肺组织湿干比增高等改变。结论所建立的输注人存储后全血中分离的血浆的TRALI大鼠模型具有可行性、实用性与稳定性等特点,为诊断和治疗TRALI提供了实验基础。 Objective To study the rat model of TRALI induced by plasma of human, and to figure out the pulmonary pathological changes. Methods The plasma of human blood whose type was AB was transfused via vein into male Sprague Dawley rats,which had been pretreated with LPS, to establish the model of TRAL/. Before the transfusion, the plasma had storaged for 21 days. The changes of pulmonary pathology, as well as the wet and dry was analyzed. Results The rat model of TRALI established successfully, whose pulmonary pathology resulting in thickening of alveolar septal and bronchial wall, alve- olar fibrin infiltration,alveolar hemorrhage, as well as the increase of wet and dry. Conclusion The rat model of TRALI could be established successfully by transfusing the plasma of human blood which had been stored. This model provides a perfeet experimental basis for the diagnosis and treatment of TRALI because of the feasibility, practicality and stability.
