摘要
采用EST-SSR标记对14个茶树新品种进行了遗传多样性分析和分子指纹鉴定。结果表明:10个SSR标记在14个品种中共检测到29个等位基因,平均每个标记2.9个,PIC和遗传多样性指数平均值分别为0.387和0.756,参试品种中多态性适中。不同等位基因的出现频率有较大差异,其变异范围在3.57%~89.29%。参试品种的Nei′s遗传距离(D)在0.036~0.472之间,当D=0.12时,可将14个茶树品种聚为三类。利用4个核心标记即可区分全部14个品种,并根据获得的等位基因带型,构建了各个品种的分子指纹图谱。
Using EST-SSR marker to analyze genetic diversity and molecular fingerprinting of 14 new tea tree varieties.The results showed that total 29 alleles were detected among 14 varieties by 10 SSR markers with average of 2.9;the average values of PIC and genetic diversity index were 0.387 and 0.756,respectively,so the tested varieties had moderate polymorphism.The occurrence frequency of different alleles had large difference with a variation range of 3.57%~89.29%.The Nei's genetic distance(D) of tested varieties ranged from 0.036 to 0.472,the 14 tea tree varieties can be gathered to three categories when D=0.12.Using four core markers can divide all of 14 varieties,and the molecular fingerprinting of each variety can be constructed according obtained allele band type.
