摘要
目的探索羟基喜树碱对乳腺癌细胞MCF-7细胞信号转导相关基因TIMP-3表达,观察羟基喜树碱处理乳腺癌细胞MCF-7细胞形态的改变,从而探讨羟基喜树碱在乳腺癌治疗中的潜在新靶点。方法以乳腺癌细胞MCF-7为研究模型,细胞暴露于羟基喜树碱浓度为10、40、80mg/L的培养液中24h,PBS组为对照组。采用免疫组化法检测羟基喜树碱(HCPT)对乳腺癌MCF-7细胞TIMP-3表达特征及细胞形态的变化。结果 HCPT对TIMP-3诱导表达作用差异不明显,但HCPT处理组细胞呈现明显的空泡化。结论 HCPT可诱导乳腺癌MCF-7细胞空泡化作用。
Objective To explore the different concentration of HCPT effects on TIMP-3 expression in breast tomor cell line MCF-7 and the shapes of MCF-7 cells were observed in this paper. Methods MCF-7 cell cultures were exposed to HCPT at concentrations of 10, 40, 80 mg/L for 24 h. PBS group as control group. Immunocytochemical method was used to measure the TIMP-3 expression in breast tomor cell line MCF-7 and the shapes of MCF-7 cells for each sample. Results The upregulated expression of TIMP-3 in the MCF-7 cell line could not be obviously induced by different concentration of HCPT, but the obvious presentation of the cellular vaculization could be induced by the cells of the disposed guoup of HCPT. Conclusion The cellular vaculization for MCF-7 cells can be induced by HCPT.
