摘要
根据番茄环斑病毒(ToRSV)、南芥菜花叶病毒(ArMV)和草莓潜隐环斑病毒(SLRSV)3种检疫性病毒的外壳蛋白基因保守序列设计特异性引物,在建立各种病毒单个RT-PCR技术的基础上,通过优化多重RT-PCR反应条件,初步建立了同步检测这3种病毒的多重RT-PCR检测体系。结果表明:3对引物特异性较强,分别可以扩增出580、438、298bp的目的片断;该方法快速、灵敏、准确,为同时检测进境百合中多种病毒提供了参考。
The specific primers were designed according to the conserved sequences of coat protein genes of three quarantine viruses-tomato ringspot virus(ToRSV),arabis mosaic virus(ArMV)and strawberry latent ringspot virus(SLRSV),and the single detection of ToRSV,ArMV and SLRSV was established by RT-PCR.Based on the above results a multiplex RT-PCR system of synchronously detecting those three viruses was preliminarily established by optimization of multiplex RT-PCR conditions.The experimental results showed that three pairs of the primers had strong specificity and could be used to amplify the 580 bp,438 bp and 298 bp target DNAs.This method was rapid,sensitive and accurate,providing a reference for synchronously detecting several kinds of viruses in introduced lily seedlings.
