摘要
目的了解胚胎十二指肠上皮的组织发生.方法光镜观察 ICR 小鼠 d 11~d 19胚胎十二指肠上皮的变化;体视学法测量胚胎 d 12~d 15十二指肠近幽门段(D_1),乳头近侧段(D_2),乳头远侧段(D_3)的上皮和肠腔面积(EA,CA)及上皮细胞,分裂细胞,凋亡细胞的面积密度(DEC,DMC,DAC).部分切片以 TUNEL 法显示凋亡细胞作对照.结果在胚胎 d 13~d 16十二指肠 D_1段上皮变化明显.胚胎d 12 EA(9.2±1.2)×10^(-3)μm^2,CA(1.2±0.5)×10^(-3)μm^2,DEC(0.015±0.003)×10^(-4)/μm^2,DMC(4.2±2.3)×10^(-4)/μm^2及 DAC(0.4±0.1)×10^(-4)/μm^2.胚胎 d 13十二指肠上皮为复层并出现上皮皱褶及次级腔;EA(21.2±6.3)×10^(-3)μm^2,CA(2.7±1.0)×10^(-3)μm^2及 DEC(0.019±0.003)×10^(-4)/μm^2均增高(P<0.01).胚胎 d 14上皮皱褶及次级腔增多,原发腔狭小呈裂隙状,DMC(2.5±1.2)×10^(-4)/μm^2和 DEC(0.017±0.002)×10^(-4)/μm^2降低(P<0.01)而 DAC(9.8±4.2)×10^(-4)/μm^2却显著增高(P<0.01).胚胎 d 15多数次级腔与原发腔相通融合,原发腔扩大,同时 DAC(8.9±4.0)×10^(-4)/μm^2降低.胚胎 d 16已见肠绒毛.十二指肠 D_2,D_3段的变化与 D_1段相似.
AIM To understand the histogenesis of duodenal epithelium of mouse fetuses. METHODS ICR mouse fetal duodenum aged 11-19 days was observed under light microscope.The epithelial and cavity area (EA,CA) and the density of epithelial,mitotic and apoptotic cells (DEC,DMC and DAC) in the near pylorus of duodenal segment (D_1),the proximal and distal segments of duodenal papilla (D_2,D_3) during 11-15 days were measured by stereologic method.Apoptotic cells in some sections were labeled by TUNEL method. RESULTS The epithelial changes were notable in D_1 segment during 13-16 days.EA was (9.2±1.2)×10^(-3) μm^2,CA (1.2±0.5)×10^(-3)μm^2,DEC (0.015±0.03)× 10^(-4)N/μm^2,DMC (4.2±2.3)×10^(-4)N/μm^2 and DAC (0.4± 0.1)×10^(-4)N/μm^2 on the 12th day.The epithelium was stratified and the epithelial folds and the secondary cavities formed and EA (21.2±6.3)×10^(-3) μm^2,CA (2.7± 1.0)×10^(-3)μm^2 and DEC (0.019±0.003)×10^(-4)N/μm^2 all increased on the 13th day (P<0.01).There were more epithelial folds and the secondary cavities,and the primary cavity was narrow or crevice and DMC (2.5±1.2) ×10^(-4)N/μm^2 and DEC (0.017±0.002)×10^(-4)N/μm^2 diminished (P<0.01) but DAC (9.8±4.2)×10^(-4)N/μm^2 increased dramatically on the 14th day (P<0.01).Most secondary cavities were fused with the primary cavity and enlarged on the 15th day but DAC (8.9±4.0)×10^(-4)N/μm^2 began to decrease,intestinal villi could be seen on the 16th day.The changes in D_2 and D_3 segments were similar to D_1 segment. CONCLUSION Histogenesis of fetal duodenal epithelium is related to epithelial cellular mitosis and apoptosis.
