摘要
对北极放线菌Streptomyces sp.MLA-21的发酵条件进行优化,提高发酵液中抗肿瘤活性部位的产量。以发酵液对肺癌A549细胞的抑制率(IR)作为观测指标,利用响应面分析法对通过单因素实验和Plackett-Burman设计筛选出培养基初始pH值、黄豆浸出物含量,可溶性淀粉含量等关键因素进行优化。建立二次回归模型。得到菌株MLA-21发酵的最优条件为KNO_31.0 g/L,可溶性淀粉23.04g/L,黄豆浸出物7.1g/L,K_2HPO_4 0.5g/L,MgSO_4 0.5g/L,NaCl 0.5g/L,pH 7.67,接种量:7.5%,培养温度:28℃,摇床转速:150 r/min。对优化结果进行验证发现优化后的培养基能够显著提高发酵液对A549细胞的抑制率和活性部位的产量。
The fermentation conditions of strain Streptorayces sp. MLA-21 were optimized to elevate the yield of the fraction with cytotoxic activity against human lung cancer cell line A549 from the fermentation broth. The inhibitory rate of the broth was selected as the measured value. The Response Surface Methodology (RSM) was applied to grade the significant factors of medium pH value, soluble starch and soybean extract filtered by Plackett-Burman design and One-factor experimental design. Based on the second order moder,the optimal conditions for strain MLA-21 were determined as follows: KNO3 1.0 g/L, soluble starch 23.04 g/L,soybean extract 7.1 g/L, K2HP04 0.5 g/L, MgSO4 0.5 g/L, NaC1 0.5 g/L, pH 7.67, inoculums size 7.5% , cultivation temperature 28 ℃ and rotation speed 150 r/min. Verification of the optimal conditions confirmed that the inhibitory rate of the broth and the yield of active fraction were both notably increased.
