摘要
目的:探讨肿瘤坏死因子α(TNFα)对血管内皮细胞(VEC)损伤的机制以及在急性肺损伤(ALI)发病中的作用。方法:通过建立人脐静脉内皮细胞(HUVEC)体外培养,采用流式细胞技术观察TNFα对HUVEC凋亡的影响以及抗TNFα的保护效应。结果:TNFα加入细胞培养中,可明显诱导HUVEC凋亡,并随着作用时间的延长和TNFα浓度增加,细胞凋亡率也相应增加,一定范围内呈时间、剂量、效应关系。而抗TNFα单克隆抗体可明显阻断由TNFα诱导的细胞凋亡。结论:细胞凋亡是VEC受损的主要方式之一。
Objective:To explore the potential mechanism underlying tumor necrosis factorα
(TNFα) on vascular endothelial cell (VEC) damage.Methods:By using cytometry technique,we
studied the effects of TNFα on apoptosis of human umbilical vein endothelial cells(HUVECs) in
vitro,and inhibitory effect of antiTNFα monoclonal antibody (McAb) on apoptosis in TNFα
induced programmed cell death (PCD).Results:TNFα was able to induce apoptosis of HUVECs
and treatment with antiTNFα McAb protected from TNFαinduced apoptosis in a time and
dosedependent fashion.Conclusions:These data suggest that apoptosis may play a role in TNF
αinduced damage of vascular endothelial cells.
