摘要
α-淀粉酶是产量大用途广的酶制剂之一。促使α-淀粉酶高产的基因是它的调节基因,而它的调节基因又有两种,一种是既可调控α-淀粉酶结构基因,又可调控蛋白酶结构基因以及其它酶结构基因;
Tunicamycin-resistant genes were map-ped near the α-amylase structure gene (amyE)and regulation gene (amyR).The gene or-der around amyE is Lin2-tmrA7-amyR-amyE-tmrB8-aroI906.Tunicamycin-resistant mu-tants were isolated from B.subtilis BF 7658after treatment with NTG.They are ableto grow normally in a BPY mediumcontaining 50 μg of tunicamycin per ml of so-lution.The mutants were classified into twotypes by activity of extracellular α-amylase.One type which is about 28% in total mutantscan raise the activities of α-amylase.Butanother type which is about 72% in total mu-tants did not change the activities though theyare tunicamycin-resistant mutants.We have screened the high productionstrains which raiseed the activity of α-amyla-se about 50% from B.subtilis BF7658.
