骨髓基质干细胞移植对大鼠脊髓损伤区NGF与BDNF表达的影响 被引量:5

Effects of bone mesenchymal stem cells transplantation on the expression of nerve growth factor and brain-derived neurotrophic factor protein in damaged zone after the spinal cord injury of rats

摘要 目的研究骨髓间充质干细胞(bone mesenchymal stem cells,BMSCs)移植对大鼠脊髓损伤(SCI)后神经生长因子(NGF)与脑源性神经营养因子(BDNF)表达的影响。方法大鼠SCI后10minBMSCs移植入损伤区,应用west-ern blot观察BMSCs移植后,大鼠脊髓损伤区NGF和BDNF蛋白表达的变化。结果各时间点上NGF、BDNF蛋白在BMSCs移植组较对照组表达明显增强。NGF表达水平在3d达高峰后开始下降,BDNF3d达高峰后到14d仍然维持在较高水平。结论脊髓损伤后BMSCs移植可上调脊髓损伤区NGF、BDNF的表达,是修复脊髓损伤的机制之一。 Objective To observe the effects of bone mesenchymal stem cells(BMSCs) transplantation on the expression of nerve growth factor(NGF) and brain-derived neurotrophic factor(BDNF) in damaged zone after the spinal cord injury(SCI) of rats.Methods 10 minutes after the operation of SCI,the BMSCs were transplanted into the damaged zone.Then NGF and BDNF protein in the damaged zone were tested by Western blot.Results Compared with control group,transplantation of BMSCs enhanced the expression of NGF and BDNF protein significantly in the damaged zone at the different stages.The expression of NGF protein peaked at 3d,then down-regulated.The expression of BDNF protein peaked at 3d,then lingered until 14d.Conclusion BMSCs transplantation can up-regulate the expression of NGF and BDNF in damaged zone after the SCI of rats.It is one of the mechanism of repairing the SCI by BMSCs transplantation.
