一个新的家蚕LIMonly蛋白基因的序列分析和原核表达

摘要 LIM蛋白是一类含有LIM结构域的真核蛋白家族,它可介导细胞分化和发育,转录调控和激活、参与细胞骨架形成等多种重要的生物调节过程。从家蚕蛹cDNA文库获得一个新的编码LIM only蛋白的基因,我们将其命名为BmLIMO(BombyxmoriLIMonly protein)。该基因ORF长度为561 bp,编码长度为186个氨基酸残基的蛋白质,预测分子量为21.6 kDa,等电点为9.14。对BmLIMO基因进行了生物信息学方面的分析,并用DNA重组技术将Bm-LIMO片段克隆到原核表达载体pET-28α中,在大肠杆菌BL21(DE3)中成功表达了可溶性BmLIMO蛋白,这为进一步研究该蛋白功能奠定了基础。 The eukaryotic LIM protein family is characterized by one or more conserved LIM domains which have been shown to be critical for the cellular differentiation and development,transcriptional control and the cytoskeleton formation.A gene encoded an LIM protein and was identified from a cDNA library of silkworm pupae.It was named BmLIMO(Bombyx mori LIM only protein).The full length of its ORF was 561 bp encoding 186 amino acids with their predicted mass 21.6 kDa and isoelectric point 9.14.After bioinformatic analyses on BmLIMO,the fragments of interest were cloned into the expression vector pET-28α and transformed into E.coli BL21(DE3) to induce protein expression.This will provide us important evidence to make further studies of BmLIMO.
